The invention relates to application of high-purity EPA-E and DHA polyenoic acid ethyl ester in preparation of nutritional supplement for patients in operative period, wherein the high-purity EPA-E and DHA polyenoic acid ethyl ester contains eicosapentaenoic acid ethyl ester and docosahexenoic acid ethyl ester with the weight percent of more than 80 percent; and the weight ratio of the EPA-E and EHA-E is 1:1.01-1:3.8. In the invention, because the total content of the EPA-E and the DHA-E is more than 80 percent, fatty acids impurities and non-fatty impurities in fish oil are greatly removed, and adverse reaction is reduced; preparation formulation is proper, the EPA-E and the DHA-E are prevented from being oxidized, the preparation formulation is taken orally and is absorbed in intestine,and the risk of infection is reduced; and the detection for the lipid level of the patients in the operative period for every day is not needed so that the treatment cost is reduced.
