Embodiment 1

[0053] 1. Preparation of explants

[0054] 1. Aseptic pre-culture

[0055] Select the seeds of Tetraena mongolica that are full of embryos (documents recording the material: Wang, G., QQLin, et al. (2007). "Tetraena mongolica Maxim can accumulate large amounts of triacylglycerol in phloem cells and xylem parenchyma of stems. " Phytochemistry 68(15): 2112-2117.; the public can obtain it from the Institute of Botany, Chinese Academy of Sciences), after soaking in sterile water with 1% Tween for 10 minutes, sterilizing with 70% ethanol for 1 minute, and soaking with 10% sodium hypochlorite for 6-7 minutes, Rinse with bacteria water 4-5 times. Inoculate the seeds on a hormone-free 1 / 2MS medium for about 10 days to form sterile seedlings.

Abstract

The invention discloses a method for breeding transgenic tetraena mongolica callus and a special culture medium thereof. The method comprises the following steps: (1) transplanting a tetraena mongolica stem into the tetraena mongolica callus induction culture medium, and culturing to obtain the callus; (2) and transforming the callus obtained in step (1) by using Agrobacterium rhizogenes containing target gene to obtain the transgenic tetraena mongolica callus. In the method, tetraena mongolica seeds from Wuhai in Inner Mongolia are selected as the initial experimental material; after sterileculture to obtain sterile seedlings, the young stem section is cut off to induce the callus; and by using the callus as the explant, and extraneous gene is integrated into the genome of tetraena mongolica by using the Agrobacterium rhizogenes mediated method. By optimizing the processes of transforming and screening callus, the invention establishes a tetraena mongolica callus transformation system which has the advantages of simple operation, high efficiency and high speed.

Description

Technical field [0001] The invention relates to a method for cultivating genetically modified tetrahedron callus and its special culture medium, belonging to the field of tetrahedron biotechnology. Background technique [0002] Tetraenarium is a small deciduous shrub of the genus Tetraenae of the Zygophyllaceae family, and it is a single species of genus unique to my country. It is a strong xerophyte. It is distributed in the northern part of the Ordos Plateau, south of the Kubuqi Desert, and the foothills of the Alabasu Mountain in the Inner Mongolia Autonomous Region. The ecological conditions such as low rocky mountains, gravel plateaus and piedmont alluvial fans are extremely harsh In the arid area, it can prevent wind and sand, conserve water and soil, and play an important role in maintaining the function of the desert ecosystem. It is the most representative ancient relict and endangered rare plant. However, due to the low seed setting rate and high embryo abortion rate, ...
