Problems solved by technology

[0006] The food samples to be inspected cannot be sterilized, because this will affect the quality of the food; aseptic technology is the basic requirement of microbiology, and this reality brings great problems to the use of microbial detection methods: The source of these interfering microorganisms is unclear, and there are many types, which seriously affect the growth of the detected bacteria (inhibit or promote the growth of the detected microorganisms), or interfere with the proliferation of microorganisms and affect the indication results of the acid-base indicator, which has a great impact on the detection results. False Negative or False Positive Results

Embodiment 1

[0036] Example 1: Isolation and identification of Bifidobacterium breve LJM-006 strain

[0037] The feces of a healthy young man in Zhejiang Province were used as the separation sample, and the Bifidobacterium breve LJM-006 of the present invention was obtained by spreading and separating culture on MRS agar medium at 37°C under anaerobic conditions for 48 hours. Bifidobacterium breve, this strain has been in the General Microbiology Center of China Microbial Culture Collection Management Committee on October 28, 2011 (Address: Institute of Microbiology, Chinese Academy of Sciences, No. 3, Beichen West Road, Chaoyang District, Beijing, 100101) Preserved, classified and named as Bifidobacterium breve (Bifidobacterium breve), and the preservation number is CGMCC No. 5418.

[0038] The formula of MRS agar medium: peptone 10g, beef extract 10g, yeast extract 5g, glucose 20g, sodium acetate 5g, K 2 HPO 4 2g, MgSO 4 ·7H 2 O 0.5g, MnSO 4 ·4H 2 O 0.2g, oligofructose 3g, diammonium citrate...

Embodiment 2

[0050] Example 2: Detection of penicillin drug residue in pig liver samples

[0051] (1) Strain activation: Inoculate the Bifidobacterium breve LJM-006 strain on nutrient agar slant medium, adjust the pH of the medium to 6.8~7.2, optimize culture at 35~37℃, and transfer Subculture

[0052] (2) Preparation of bacterial suspension: Take the Bifidobacterium breve LJM-006 obtained in step (1) and inoculate it on nutrient agar slant medium, cultivate it anaerobicly at 35±2°C for 24 hours, and wash the lawn with sterile normal saline. Make a suspension and adjust its OD 600 Value to 0.25~0.35, store in refrigerator at 2-8℃;

[0053] (3) Preparation of test tube: Take 10g peptone, 10g beef extract, 5g yeast extract, 20g glucose, 5g sodium acetate, K 2 HPO 4 2g, MgSO 4 ·7H 2 O 0.5g, MnSO 4 ·4H 2 O 0.2g, oligofructose 3g, diammonium citrate 2g, Tween-80 1mL, distilled water 1L, heated to dissolve to correct pH to 6.5, autoclaved at 115°C for 15-20 minutes, cooling temperature at 50-65°C, A...

Embodiment 3

[0068] Example 3 Detection of other samples

[0069] Meat, liver, kidney and other tissues can be weighed and squeezed to extract the juice and add an appropriate amount of phosphate buffer to adjust the pH to 7.0; for different animal samples, add different buffers to control the pH. Other operation steps are the same as in Example 2.

Abstract

The invention discloses a test method for rapidly screening antibacterial residue in animal food samples, which comprises the steps of: strain activation, bacterial suspension preparation, test tube preparation, sample preprocessing and sample testing. The test method utilizes the principle that antibacterial can inhibit the growth or reproduction of bacteria sensitive to the antibacterial, a sample to be tested is sucked and added into a test tube, and is cultured for a certain period of time, and according to the change of the color of the culture medium in the test tube, whether the sample contains antibacterial residue or not is judged. Compared with the prior art, the test method keeps the reliability of the TTC (triphenyltetrazolium chloride) test method, and solves the problem that in the TTC test method, the interference of autochthonous microorganisms or miscellaneous bacteria in the food sample leads to false positive and false negative results; and the test method has the characteristics of simplicity, little interference, high sensitivity, high accuracy and the like, and is applicable to the high-throughput rapid screening of antibacterial residue in the animal food samples.

Description

Technical field [0001] The invention relates to the technical field of detection and analysis of antibacterial drug residues, in particular to a detection method for rapidly screening penicillin drug residues in food samples by using the growth of bacteria to be inhibited by antibacterial drugs in tested samples. Background technique [0002] Antibacterial drug residues have always been a serious problem plagued by food safety. Penicillin drugs are an important class of β-lactam antibiotics. Because of their high efficiency and low toxicity, they are currently one of the most popular drugs in clinical veterinary medicine. Due to their widespread use, they are used in animal-derived foods. The residual phenomenon on the above is getting more and more serious. Although penicillin drugs themselves are not very toxic to the body, they are easy to cause drug allergic reactions. They are the most allergic drugs at the same time. If long-term consumption of low concentrations of such an...
