The method includes the steps of firstly, preparing ozone by an ozone generator, and removing nitrogen oxides; secondly, heating and grinding samples to be detected, and selecting samples to be sent into a reaction chamber; thirdly, fully performing reaction to ozone and the samples when reaction starts, and detecting a luminescent signal by a photomultiplier; fourthly, converting an optical signal into an electric signal, sending the electric signal into a data processing system of a computer, and analyzing a luminescent value-luminescent time power function and luminescent time; and fifthly, estimating lipid content according to the luminescent value-luminescent time power function per unit mass, and estimating contents of crude cellulose and crude lignin according to the luminescent time per unit mass.
