Problems solved by technology

[0006] However, if this method is used to separate and enrich flavonoid aglycones, there are many problems
We found that it is difficult to use the traditional polyamide separation and enrichment process. The reasons are: first, the flavonoid aglycone to be separated is relatively fat-soluble, and cannot be dissolved by distilled water; second, although it can be diluted with an appropriate amount Ethanol is dissolved, and then diluted with a large amount of distilled water to dissolve, but its volume will be very large; third, polyamide has the strongest adsorption capacity for flavonoids in aqueous solution, will the diluted ethanol sample solution affect the adsorption of flavonoid aglycone still to be verified
In practice, no matter we use polyamide to separate and enrich flavonoid aglycones or flavonoid glycosides, we have similar problems. However, after searching the literature, there is no relevant literature discussing this problem, and almost all literatures only isolate and enrich the total flavonoids

Abstract

The invention discloses a method for extraction of total flavonoid aglycones from hickory leaves. The method comprises: subjecting hickory leaf dry powder to heating reflux extraction by an ethanol aqueous solution with a volume concentration of 50-95%, conducting filtering to obtain a total flavonoid aglycone extract solution; conducting a pretreatment on polyamide; adding the pretreated polyamide into the total flavonoid aglycone extract solution, adding water and performing vacuum distillation to obtain a concentrated mixed solution, directly applying the mixed solution on a column, and letting the solid polyamide settle naturally to a smooth and bubble-free state; releasing the liquid from the column, then first employing an ethanol aqueous solution with a volume concentration of 20-30% to wash away impurities, then using an ethanol aqueous solution with a volume concentration of 40-50% as an eluent to perform elution, collecting the eluent, and evaporating the eluent, thus obtaining the total flavone glycosides. The total flavone glycosides obtained by the method provided in the invention has the advantages of: high purity and high recovery rate. In addition to simple process and convenient operation, the method also has a low cost, and is suitable for industrialized production.

Description

technical field [0001] The invention relates to a method for separating and purifying active ingredients of traditional Chinese medicine by using polyamide, more specifically, a new method for separating and enriching total flavonoid aglycone in pecan leaves with polyamide, and belongs to the technical field of separation and purification of traditional Chinese medicine chemistry. Background technique [0002] Flavonoids are naturally occurring organic compounds. In nature, flavonoids are widely found in various plant organs (such as seeds, flowers, fruits, and leaves) and other Chinese medicinal materials, such as Huixincao, sweet tea, nasturtium, motherwort, ginkgo biloba, hawthorn, Sugarcane, chestnut flowers, propolis, etc. are rich in flavonoids. A class of compounds formed by connecting two benzene rings (A ring and B ring) through a three-carbon chain are collectively called flavonoids. Substituted by group, isopentenyl, etc. According to the connection position of...
