Embodiment 1

[0023] Identification of the 16SrRNA molecule of the strain

[0024] The actinomycete strain JSX-1 was activated and cultured on Gaoshi No. 1 plate at a temperature of 28°C. Gaoshi No. 1 medium: soluble starch 19.9-20.1g, sodium chloride 0.4-0.6g, potassium nitrate 0.9- 1.1g, dipotassium hydrogen phosphate 0.4-0.6g, magnesium sulfate 0.4-0.6g, ferrous sulfate 0.009-0.011g, agar 15-20g, pH 7.1-7.3, dilute the above components to 1000ml with distilled water, Sterilize at a temperature of 120-121°C for 19-21 minutes.

[0025] After 2-4 days of culture, it was found that the single colony of JSX-1 was round, white, and the surface was dry and opaque; the mycelium was thin, the growth was slow, and the colony was dense in texture and had a muddy smell. Strain JSX-1 Gram stain was purple and positive.

[0026] The morphological characteristics of the strain JSX-1 were most similar to those of Streptomyces sp. in the identification manual of common bacterial systems.

Embodiment 2

[0030] The preparation method steps of Phytophthora soybean zoospore suspension are as follows:

[0031] (1) Add CaCO3 to V8 vegetable juice at a ratio of 1:100 (w / v), then centrifuge at a speed of 6000-8000r / min for 8-10min, discard the bottom sediment, and add 9 times the volume of distilled water is prepared into 10% V8 vegetable juice medium, and the solid medium is added to the agar at a ratio of 1.5-1.8%. The temperature is 120-121°C, and the sterilization time is 15-20min.

[0032] (2) The preserved Phytophthora sojae strain was activated and grown on the V8 medium at a temperature of 24-26° C., placed in the dark in a constant temperature incubator for cultivation, and the activated growth time was 5-6 days.

[0033] (3) Cut out the Phytophthora sojae strain with a scalpel of 2-3mm×2-3mm in size from the above-mentioned Phytophthora sojae strain cultured for 5-6d.

[0034] (4) Put 10-20 pieces of the above-mentioned hyphae into 14-16ml of 10% liquid V8 vegetable juic...

Embodiment 3

[0056] Determination of control effect of detached leaves

[0057] Cultivate the actinomycete bacterium until the predetermined time for use; the bacterium can be used after irrigating the roots 2-3 days after emergence. Collect healthy leaves of the same size from neatly grown soybean plants, soak the soybean leaves with 0.5% Tween 20 for about 9-10 seconds, rinse gently with sterile water, take them out and dry them in an ultra-clean workbench. After drying the leaves, soak them in the actinomycetes agent for 1 min, take them out and dry them in the ultra-clean workbench (leaves soaked in the supernatant of the actinomycetes fermentation medium are used as a control), and inoculate 4-6 μl of Phytophthora sojae on the leaves For zoospore suspension, each treatment was repeated 6-8 times. At 24-26° C., relative humidity 80-90%, and dark conditions for 48 hours, measure the diameter of the lesion and evaluate the control effect. The formula for calculating the control effect ...

Abstract

The invention discloses an actinomycete strain JSX-1 for preventing and curing phytophthora sojae, and belongs to the technical field of plant protection. The strain belongs to streptomycete (Streptomyces sp.) and is named as actinomycete JSX-1. The single colony of the actinomycete strain is circular, white and dry and non-transparent in surface; the hypha is thinner, and grows slowly; the bacterial colony is compact in texture and has a fishy smell. The gram stain of the actinomycete strain is purple, and positive. The JSX-1 fungicide has the controlling effect of 55 to 60 percent on phytophthora sojae. The invention provides the actinomycete strain JSX-1 for preventing and curing phytophthora sojae and the fungicide thereof which are not liable to enable phytophthora sojae to produce resistance, and has no pollution to environment, and the fungicide can be used through root-irrigation after soybean seedling emergence.

Description

technical field [0001] The invention relates to the field of microorganisms, in particular to an actinomycete strain JSX-1 for preventing and treating soybean phytophthora and its agent. Background technique [0002] Soybean is one of the most important crops in agricultural production. Phytophthora soybean caused by Phytophthorasojae is a worldwide disease that endangers soybean production. The disease is an extremely serious soil-borne disease, and it is also an important foreign quarantine object in my country. Soybean Phytophthora was first discovered in Indiana, USA in 1948, and then gradually occurred in the United States and many countries in the world. It was not until 1989 in my country that Shen Chongyao first isolated Phytophthora sojae in the northeast soybean production area. Currently, the disease is affecting some soybeans in my country. Production areas have also occurred and caused serious harm. [0003] Phytophthora soybean can survive in the soil for a lo...
