The invention relates to extracting phytosterol from pulp floating oil pitch and a preparation method of sterol ester, which comprises the following steps of: 1, obtaining black liquor by a paper making process, separating insoluble materials, performing evaporation and concentrating, filtering, extracting and then extracting an upper liquid layer to obtain a black soap layer; 2, preparing an H3PO4 solution, adding black soap liquor prepared in the step1 under the stirring action, introducing the hot air into the H3PO4 solution in a reaction process; washing the obtained useful material in air with hot water, and performing precipitation to obtain the prepared crude plant floating oil; performing reduced pressure distillation to obtain floating oil pitch; 3, purifying the floating oil pitch; 4, adding 3-methyl-1,3-butanediol, 1,4-butanediol, potassium hydroxide, calcium hydroxide and lithium hydroxide into the floating oil pitch obtained in the step 3, reacting to remove excess solvents to obtain a mixture; 5, adding trimethylpentane and dioxane into the mixture obtained in the step 4, separating, and then adding pyridine into the mixture for crystallization so as to obtain phytosterol, and thus obtaining the plant sterol ester under the action of enzyme catalysis.
