Problems solved by technology

[0006] In view of the above-mentioned technical problems, the present invention provides a method for excavating and selecting biosynthetic metabolic markers of microalgae, using three close relatives of Chlorella freshwater, Nannochloropsis seawater, and Coccus polaris under the conditions of nitrogen deficiency induction. Algae strains were used as the subjects, and based on cytoomics, metabolomics, and bioinformatics methods, three closely related algal strains were excavated and selected to synthesize and accumulate the same metabolic markers in cells. Complementary analysis can effectively avoid the one-sidedness of the results caused by only a single algae strain and a single analysis method, thereby greatly increasing the selection and identification rate and corresponding accuracy of key metabolic markers for microalgae oil synthesis and accumulation. Provide a theoretical basis for the common mechanism of microalgae oil synthesis and metabolism

Embodiment

[0026] 1. Nitrogen-deficiency conditions induce the accumulation of lipids in freshwater Chlorella, seawater Nannochloropsis, and polar coccus cells

[0027] Pick freshwater Chlorella, seawater Nannochloropsis, and Polar Coccus algae that were preserved on the plane and insert them into 30mL of the medium (A-1), (B-1), and (C-1) described below, respectively. Under the conditions of 28°C (Chlorella), 25°C (Nannochloropsis), and 27°C (Gylococcus) in dark and light alternately (12 hours: 12 hours), cultivate for 120 hours to obtain activated algae seed liquid, pipette 5 mL of activated seed solution was inserted into 100 mL of fresh (A-1), (B-1), (C-1) medium as described below, and cultivated under the condition of 25~28 ℃ (the photoperiod ratio was 12 hours: 12 hours) 5 to 10 days. Centrifuge to collect cultured algae liquid, and immediately transfer to fresh nitrogen-free Basal basal medium (A-2), nitrogen-free artificially simulated seawater basal medium (B-2) and nitrogen-...

Abstract

The invention discloses a method for excavating and selecting anabolic markers of microalgae oil. In a nitrogen deficiency induction condition, three kinds of algae with extremely poor affinis, including freshwater chlorella, seawater nannochloropsis oculata, and polar coccomyxaceae, are used as subjects, and based on cytomics, metabonomics, andbioinformatics methods, a same anabolic marker synthesized and accumulated by the three kinds of algae cell oil with extremely poor affinis is excavated and selected. Using complementary analysis among different algaes and different omics, a result one-sidedness caused by using a single algae and a single analysis method can be effectively prevented, so as to greatly increase selection identification rate of a key anabolic marker synthesized and accumulated by the microalgae oil and corresponding precision. The method provides theoretical basis for further improving yield of the microalgae oil and revealing an anabolism general mechanism of themicroalgae oil.

Description

technical field [0001] The invention belongs to the field of bioanalytical chemistry, and in particular relates to a method for excavating and selecting biosynthetic metabolic markers of microalgae oil. Background technique [0002] In recent years, the problems of energy shortage and environmental pollution have become increasingly prominent, which have restricted the development of the global economy to a certain extent, and the development of environmentally friendly renewable energy is imminent. Microalgae are considered to be one of the most potential raw materials for the development of biodiesel because of their advantages such as high photosynthetic efficiency, fast growth rate, no occupation of cultivated land, high oil content, and strong environmental adaptability. However, the cell density per unit volume of many energy microalgae is negatively correlated with the oil content, resulting in unsatisfactory oil production efficiency of the algae cells themselves, an...
