Abstract

The invention discloses a method for changing the morphological characteristics of the stems, the leaves and the flowers of a plant, and application, and relates to the technical field of plant cultivation. The method disclosed by the invention comprises the following steps of cloning a gene BjuB033714 in Brassica juncea var. tumidaTsen etLee, and constructing an overexpression vector which takes CAMV35S as a promoter by taking pTF101 as a target vector and taking BamH I and Sma I as restriction enzyme cutting sites; taking the whole genome cDNA of the Brassica juncea var. tumidaTsen etLee as a template, and amplifying the CDS sequence of BjuB033714 by adopting a PCR method; carrying out electrophoresis on the amplified BjuB033714 gene, and amplifying a product fragment by utilizing the recovered BjuB033714 gene; carrying out multiplication culture on a single colony which is positive in PCR amplification by using an LB liquid culture medium of 100mu g / mL spectinomycin, and extracting plasmids; and performing double enzyme digestion on the extracted plasmids by using restriction enzymes Sma I and BamH I to identify whether the vector is successfully constructed or not.
