[0028] Subarachnoid hemorrhage is a common hemorrhagic cerebrovascular disease with high mortality and disability. Tumor necrosis factor receptor-associated protein 1 (TRAP1) in mitochondria plays a role in metabolic mode switching of aerobic oxidation and glycolysis. Acetylation is an important post-translational modification process. However, it remains unclear how TRAP1 and acetylated TRAP1 regulate energy metabolism after SAH. Berberine hydrochloride, an isoquinoline alkaloid in traditional Chinese medicine, regulates the expression of sirtuins 3 (SIRT3). In our experiments, we performed western blotting, metabolite assays, TUNEL staining, and behavioral tests to explore the metabolic mode switch after subarachnoid hemorrhage and the effect of TRAP1 acetylation after berberine hydrochloride intervention.

Abstract

The invention discloses application of berberine hydrochloride in improvement of early brain injury after subarachnoid hemorrhage. According to the invention, western blotting, metabolite determination, TUNEL staining and behavior testing are carried out to research the energy metabolism process after subarachnoid hemorrhage. It is speculated that berberine hydrochloride can promote deacetylation of TRAP1 protein by increasing mitochondrial deacetylase SIRT3; the berberine hydrochloride promotes glycolysis of neurons, provides energy for the neurons, increases the activity of succinate dehydrogenase and reduces active oxygen after subarachnoid hemorrhage.
