Problems solved by technology

Traditional fermentation methods usually use glucose as a carbon source, but the price of glucose is high, and the nutrient substance of glucose is single, resulting in high fermentation cost and low protein content
Molasses replaces some glucose as a carbon source, which can reduce the cost of fermentation, and molasses is rich in various trace elements, which can supplement the consumption of trace elements during the fermentation process. At present, there is still a lack of molasses combined with trace elements to optimize the fermentation parameters of Schizochytrium sp. reported that further research in this area is urgently needed

Embodiment 1

[0034] The method for cultivating Schizochytrium with high protein content by fermenting molasses comprises the following steps:

[0035] (1) Put soybean molasses into a container, and bathe in a water bath at 90°C for 1 hour;

[0036] (2) Place the molasses solution after the water bath in a centrifuge, centrifuge at 6000r / min for 10min, and collect the supernatant for later use.

[0037] (3) Take 60ml of centrifuged supernatant and add it to a 250ml shake flask, add 0.4g peptone, 0.4g yeast extract, 2g sea crystal, 0.2g defoamer, add 100ml distilled water to dissolve and sterilize for later use.

[0038] (4) Inoculate Schizochytrium sp. into the sterilized culture medium at an inoculation amount of 5%, and grow in a fermentation shaker for 96 hours.

[0039] (5) Place the cultured Schizochytrium in a centrifuge, pour off the supernatant after centrifugation, and dry the lower sediment. It has been determined that the protein content in Schizochytrium is about 30%.

Embodiment 2

[0041] (1) Put soybean molasses into a container, and bathe in a water bath at 90°C for 1 hour;

[0042] (2) Place the molasses solution after the water bath in a centrifuge, centrifuge at 6000r / min for 10min, and collect the supernatant for later use.

[0043] (3) Take 48ml of centrifuged supernatant and add it to a 250ml shaker flask, add 0.4g peptone, 0.4g yeast extract, 2g sea crystal, 0.2g defoamer, add 100ml distilled water to dissolve and sterilize for later use.

[0044] (4) Inoculate Schizochytrium sp. into the sterilized culture medium at an inoculation amount of 5%, and grow in a fermentation shaker for 96 hours.

[0045] (5) Place the cultured Schizochytrium in a centrifuge, pour off the supernatant after centrifugation, and dry the lower sediment. It has been determined that the protein content in Schizochytrium is 28%.

Abstract

The invention belongs to the field of fermentation of schizochytrium limacinum, and particularly relates to a method for producing high-protein-content schizochytrium limacinum through fermentation by using an allogenic material as a substrate. The method mainly comprises the following steps: 1) placing soybean molasses in a container, and heating in a water bath kettle at 80-100 DEG C for 0.5-1.5 h; (2) after the water bath is completed, putting the soybean molasses into a centrifugal machine, carrying out centrifugal precipitation for 5-20 minutes at 4000-8000 rpm, and collecting molasses supernate; (3) adding the molasses supernate into a container, respectively adding peptone, yeast extract, sea crystal and a defoaming agent into the container, dissolving and sterilizing for later use; and (4) inoculating schizochytrium limacinum into the prepared culture medium according to the inoculum size of 4-6%, and carrying out fermentation culture for 72-120 hours. The fermentation method is simple in process, low in pollution, high in protein content, low in cost and suitable for large-scale industrial production.

Description

technical field [0001] The invention belongs to the field of fermentation of Schizochytrium sp., and in particular relates to a method for fermenting Schizochytrium sp. with high protein content by using exogenous substances as substrates. Background technique [0002] As a traditional large agricultural country, our country has a large number of livestock and poultry, so a large amount of feed is needed to provide nutrition for animals. As an important part of the animal body, protein needs to be ingested from the outside world. Studies have shown that Schizochytrium is composed of several substances such as protein and lipid, among which the content of protein accounts for about 15%, and by optimizing the fermentation method, Schizochytrium with higher protein content can be obtained. [0003] At present, there are few types of conventional protein feeds on the market in our country, and the cost is relatively high, so a low-priced protein feed substitute is needed. Trad...
