Problems solved by technology

[0004] Ferrous glycinate is an amino acid iron chelate. The small molecule glycine group protects ferrous ions and can maintain the integrity of the molecule. It is usually used as an ideal iron nutritional supplement for the treatment of iron deficiency anemia. But there is no report so far that ferrous glycinate is used in the treatment and prevention of rheumatoid arthritis

Embodiment 1

[0052] Example 1 In vitro protective effect of ferrous glycinate on Raw264.7 cell inflammatory injury model induced by LPS and IFN-γ co-stimulation

[0053] 1. Raw264.7 cell culture

[0054] After the Raw264.7 cells were recovered, they were cultured in a petri dish with a diameter of 10 mm, and the medium was DMEM medium containing 10% fetal bovine serum (DMEM is a medium containing various amino acids and glucose, which was purchased from Thermo Fisher Scientific), the petri dish was placed at 37°C and 5% CO 2 Cultured in a constant temperature incubator, the medium was replaced every two days, and the cells in the culture dish could be subcultured when the density reached about 80%. Follow-up experiments were carried out after culturing for 3 to 5 generations according to conventional culture techniques in the art.

[0055] 2. Experimental analysis

[0056] Detection of cell proliferation by MTT (3-(4,5-dimethylthiazole-2)-2,5-diphenyltetrazolium bromide)

[0057] (1) Co...

Embodiment 2

[0082] Example 2 The effect of ferrous glycinate on the migration of RA-FLS in rheumatoid arthritis in vitro

[0083] 1. RA-FLS culture:

[0084] After resuscitation of RA-FLS, it was added to a 25T culture flask in DMEM medium containing 10% Australian fetal bovine serum (purchased from Thermo Fisher Scientific, USA), and placed at 37°C and 5% CO. 2 The cells were cultured in a constant temperature incubator, and the medium was replaced every 2 days. When the cell density reached about 95%, the cells were passaged, and the experiments were carried out within 3 to 10 passages.

[0085] 2. Scratch test:

[0086] After the cells reached an appropriate density (about 80% confluence), they were digested with trypsin for 2 minutes, and the digestion was terminated with complete medium (DMEM medium containing 10% Australian fetal bovine serum), centrifuged at 800 rpm for 5 minutes, and the supernatant was removed. After that, the cells were suspended in complete medium (DMEM mediu...

Embodiment 3

[0089] Example 3 The effect of ferrous glycinate on the secretion of inflammatory factors in M1-type macrophages induced by THP-1 in vitro

[0090] 1. THP-1 culture

[0091] After the THP-1 cells were recovered, they were placed in a 25T culture flask containing 10% fetal bovine serum in 1640 medium (purchased from Thermo Fisher Scientific, USA), at 37°C and 5% CO. 2 Culture in a constant temperature incubator, change the medium every 2 days, and cultivate to 8 × 10 5 Cells / mL can be passaged.

[0092] 2. Western blot experiment

[0093] (1) Collect the cell suspension of the above cultured THP-1 cells and inoculate them in a six-well plate at a density of 1×10 6 cells / well, 37°C and 5% CO 2 The experiment was carried out after culturing for 24 h under the conditions. The experiment was divided into six groups, namely blank control group, stimulation control group and drug-adding group of different concentrations of ferrous glycinate (4 groups), with 2 duplicate wells in ...

Abstract

The invention discloses an application of ferrous glycinate in treatment of rheumatoid arthritis. Experiments prove that after LPS and IFN-gamma jointly stimulate Raw264.7 cells, the cell activity can be remarkably inhibited, ferrous glycinate can remarkably recover the activity of the Raw264.7 cells and has a protection effect on the Raw264.7 cells, in addition, ferrous glycinate can reduce expression of inflammatory factors, inhibit expression of iNOS protein and inhibit excessive oxidative stress reaction in rheumatoid arthritis, and the effect of preventing and treating rheumatoid arthritis is achieved. And the expression of the antioxidant related protein NQO-1 is up-regulated. In addition, ferrous glycinate can also significantly inhibit migration of RA-FLS, can improve swelling and damage of mouse soles and ankle joints, can reduce levels of IL-6 and IL-1beta of inflammatory factors in mouse serum, has a significant treatment effect in a CIA model mouse body, and has good practical popularization and application values.

Description

technical field [0001] The invention belongs to the technical field of medicine, and particularly relates to the application of ferrous glycinate in the treatment of rheumatoid arthritis. Background technique [0002] Rheumatoid arthritis is a chronic autoimmune disease characterized by aggressive joint inflammation and irreversible damage to cartilage, causing symmetrical joint pain and widespread inflammatory damage to connective tissue throughout the body. The global prevalence of rheumatoid arthritis is 5 in 1,000. At present, the specific etiology of rheumatoid arthritis is not clear, and both genetic and environmental factors may lead to the occurrence of the disease. The pathological feature of rheumatoid arthritis is that a large number of inflammatory cells such as T cells, B cells and macrophages infiltrate the synovium of multiple joints. At the same time, the secretion of various rheumatoid inflammatory factors can cause joint pain, swelling, deformity and loss o...
