The invention discloses a method for increasing content of camptothecin in camptotheca hairy root in the technical field of biology. The method comprises the following steps of: cloning coding frame sequences of CrORCA3 and CrG10H genes from catharanthus roseus; constructing a plant bivalent high-efficiency expression vector including the genes; and genetically transforming the camptotheca into camptotheca hairy root genetically modified by CrORCA3 and CrG10H genes, by using the hair root agrobacterium-mediated technique. The camptothecin content in the obtained genetically-modified camptotheca hairy root is increased obviously. The content of the camptothecin is 5.4 times of the contrast. The content of hydroxycamptothecin is 2.3 times of the contract. The content of the hairy root camptothecin independently genetically-modified by CrORCA3 is increased by 1.5 times of the contract. The content of the hairy root camptothecin independently genetically-modified by CrG10H gene is not increased obviously. The invention provides a method for increasing content of the camptothecin in the camptotheca hairy root, thereby providing a novel method for producing the camptothecin and the hydroxycamptothecin as anti-cancer drugs of important clinical requirements.

The invention relates to a method to extract the vinblastine extracted from the catharanthus roseus, including the following processes: (1) the catharanthus roseus is agitated and smashed after the wetting by the acidic water and then is soaked in the acidic water, the filtrate after the filtration is regulated to alkalinity by using alkali, a alkaline water phase is extracted by using chloroform, a chloroform phase is collected, and the solid reclaimed materials can be obtained after the reclaiming of the chloroform; (2) the reclaimed materials are dissolved by using the acidic water, the ethyl acetate is added for extraction, and a acidic water phase is reclaimed; (3) the reclaimed acidic water phase in step (2) is regulated to alkalinity, the chloroform is used for extraction, and the crude alkaloid is obtained by the reclaim of chloroform of the extraction phase; (4) the obtained crude alkaloid in step (3) is dissolved by using the acidic water, of which the pH is regulated to alkalinity, the lower layer of sediments is collected centrifugally, the refined catharanthus roseus alkaloid is obtained after the drying of the sediments, and the vinblastine and leurocristine can be obtained by alumina column chromatography separation; (5) the filtrate which is obtained by centrifugation in step (4) is extracted by using chloroform, the catharanthine and the vindoline are obtained by vacuum concentration and macroporous resin separation. The method can improve the utilization ratio of catharanthus roseus.

The preparation method provided by the invention is a novel method for producing an anti-cancer drug that is camptothecin under important clinical demand.

Noscapine analogs and their use in treating cancers, including drug-resistant cancers

Compounds, pharmaceutical compositions including the compounds, and methods of preparation and use thereof are disclosed. The compounds are noscapine analogs. The compounds and compositions can be used to treat and/or prevent a wide variety of cancers, including drug resistant cancers. While the antitussive plant alkaloid, noscapine, binds tubulin, displays anticancer activity, and has a safe pharmacological profile in humans, structure-function analyses pointed to a proton at position 9 of the isoquinoline ring that can be modified without compromising tubulin binding activity. Noscapine analogs with various functional moieties at position 9 on the isoquinoline ring kill human cancer cells resistant to other anti-microtubule agents, such as vincas and taxanes. Representative analogs include the 9-nitro, 9-bromo-, 9-iodo-, and 9-fluoro-noscapines, which bind tubulin and induce apoptosis selectively in tumor cells (ovarian and T-cell lymphoma) resistant to paclitaxel, vinblastine and teniposide. Surprisingly, treatment with one of the analogs, 9-nitro-nos, at doses as high as 100 μM, did not affect the cell cycle profile of normal human fibroblasts. This selectivity for cancer cells represents a unique edge over the other available antimitotics. The compounds can perturb the progression of cell cycle by mitotic arrest, followed by apoptotic cell death associated with increased caspase-3 activation and appearance of TUNEL-positive cells. Thus, the compounds are novel therapeutic agents for a variety of cancers, including ovarian and T-cell lymphoma cancers, even those that have become drug-resistant to currently available chemotherapeutic drugs.
