Problems solved by technology

At present, formalin or 4% paraformaldehyde is often used for pathological fixation of the lungs to directly soak and fix the isolated animal lungs. This fixation method causes the lung volume to shrink sharply, compresses the alveoli and vascular lumen, and affects the peripheral bronchi and Accurate judgment o

Embodiment 2

[0051] The difference from Example 1 is that the isolated lungs of mice are adopted, the fixative is 0.6ml of 4% (mass percentage concentration) formaldehyde solution, the height between the liquid level of 4% formaldehyde solution and the level where the mouse lungs are located The distance is 6cm. The whole lungs of the mice were completely immersed in 4% formaldehyde solution for 48 h.

[0052] test results :from Figure 2A and Figure 2B As shown, after HE staining of mouse lung tissue fixed by the present invention, the tissue structure is complete and clear, the structure of the alveolar cavity is clear, the gas-blood barrier structure at the end can be seen, the small artery and small vein at the end of the lung have a clear structure, and a single layer of endothelial cells can be seen and smooth muscle cell layer; after HE staining of the mouse lung tissue fixed by the existing common method, the structure of the end of the lung is clearer, the alveoli are shrunk, t...

Abstract

The invention discloses a method for pathologically fixing an experimental animal lung, which comprises the following steps of: (1) filling a fixing liquid, namely, suspending a filling container filled with the fixing liquid onto an isolated animal lung, inserting the liquid outlet end of a filling tube connected with the filling container into trachea of the isolated animal lung, and filling the fixing liquid into pulmonary alveoli of the animal lung by utilizing the hydrostatic pressure formed by the height difference between the liquid surface of the fixing liquid and the horizontal surface of the animal lung so as to naturally extend the animal lung; and (2) soaking the fixing liquid, namely, completely soaking the animal lung obtained in the step (1) into the fixing liquid to be fixed. The method is convenient to operate, low in cost and fast in fixing; the method can be controlled in a normalizing manner; and the method can enable the animal lung to be completely, uniformly and naturally filled, so that natural shapes of lung tissues is favorably stored and the need of observing pathological change of small blood vessels and small tracheas of the lung is met.

Description

technical field [0001] The invention relates to a pathological fixation method for experimental animal lungs, which belongs to the field of biotechnology. Background technique [0002] Animal experiments are one of the most important and commonly used scientific research methods in current scientific research, and are the basic research methods of various disciplines such as pharmacology, physiology, and pathophysiology. Respiratory diseases (such as COPD, pulmonary vascular disease, lung cancer, etc.) are major diseases that endanger human health. Therefore, research on respiratory diseases is an important research content for many medical researchers. Animal models of respiratory diseases have been widely used in the pathogenesis of respiratory diseases Mechanism research and new drug development, in which the pathological results of experimental animals are important indicators for judging the success of the model and scientific research. Standardized pathological proces...
