The method comprises the following steps: adding a penicillin V internal label solution and then acetonitrile into homogenate eggs, mixing for direct extraction, centrifuging, removing the supernatant fluid, repeatedly extracting for 1-2 times, and merging the supernatant fluid to obtain an extract; adding 1ml ammonium acetate solution (pH 6.7) into the extract, placing the extract on a vortex mixer for mixing, adding methylene chloride saturated through ultrapure water, fully oscillating, mixing and centrifuging, moving medicine to the upper-layer water phase through the liquid-liquid extraction procedure, removing the upper-layer water phase, repeatedly extracting for 1-2 times, and then merging with the water phase; and finally drying through a pressure blowing concentrator.
