According to the method, the mixed fatty acid of the trichosanthes seed oil is taken as the raw material, and the polyunsaturated fatty acid (PUFA) of the mixed fatty acid is gathered and purified through a urea adduction method. The purification method sequentially comprises the following steps of preparing a urea-methanol solution, mixing the mixed fatty acid and the urea-methanol solution, adducting, leaching and separating, collecting filter liquor, extracting an organic phase, washing, drying, concentrating through decompression, and obtaining a PUFA product. The method is simple and reliable, is low in cost and has less environment contamination, and the solvent can be recycled. The rate of finished products can reach 40.92%, the purity of the finished products can reach 94.72%, and the finished products mainly comprise linoleic acid and linolenic acid.
