The purification method sequentially comprises the following steps of preparing a urea-methanol solution, mixing the mixed fatty acid and the urea-methanol solution, adducting, leaching and separating, collecting filter liquor, extracting an organic phase, washing, drying, concentrating through decompression, and obtaining a PUFA product. The method is simple and reliable, is low in cost and has less environment contamination, and the solvent can be recycled.
