The preparation method comprises the following steps: mixing sesbania powder and oyster shell powder with water to obtain sesbania-oyster shell suspension liquid; then mixing the obtained suspension liquid with a chitosan water solution to obtain a chitosan-sesbania-oyster shell composite liquor; then mixing the obtained chitosan-sesbania-oyster shell composite liquor with a dispersing agent, an emulsifier and an oil phase to obtain an emulsion containing water in oil droplets; and finally mixing the obtained emulsion with a glutaraldehyde saturated methylbenzene liquor to perform crosslinking reaction, thus obtaining the chitosan-oyster shell composite microsphere. According to the invention, sesbania powder is uniformly distributed to the surface of oyster shell powder, so as to play the roles of the thickening agent and a surface modifier and promote compounding of the oyster shell powder and chitosan; the mixing of the suspension liquid and the chitosan water solution can ensure forming of a homogeneous phase composite material, so as to promote smooth procedure of subsequent interlinking reaction and further improve the modification capability of the obtained composite microsphere to the water body.
