[0021] Experimental example 1 Isolation and taxonomic identification of Schizophyllum commune FPYF3010

[0022] 1. Isolation and Identification Methods

[0023] Remove the epidermis of the collected poplar leaves and cut them into 2mm 2 The size of the tissue block, with 2% sodium hypochlorite and 75% ethanol to disinfect the surface of the tissue, put the sterilized tissue block into sterile water and inoculate it into the water agar medium prepared in advance; the isolated strain Carry out purification culture on PDA medium, and the purified strains are preserved for strains; cut the culture medium with hyphae into 5mm 2 Then put them into a refrigerated tube filled with sterile water that has been sterilized three times by autoclaving, seal it, and store it at 4°C.

[0024] Use the improved CTAB method to extract the DNA of the strain: scrape off the mycelium on the culture medium, put it into a sterile 1.5mL centrifuge tube, put it in a refrigerated tube at -20°C, and th...

[0032] 1. Analysis method

[0033] The strains of Schizophyllum commune FPYF3010 were inoculated on the PDA medium, and each strain was repeated three times; placed in a constant temperature incubator for 5 days.

[0034] GC-MS detects volatile gases, and uses HP-5MS type capillary column (coating: (5% phenyl)-methylpolysiloxane, 30.0m × 0.25mm, coating thickness: 0.25μm) to separate volatile gases Gas, nitrogen as carrier gas. The heating program of the capillary column is as follows: maintain 33°C for 2min, then raise the temperature to 220°C at a rate of 7°C / min and maintain for 7min. Mass spectrometry scan rate: 5.0000scans / s. The range of mass spectrum is 41-560amu (atomic mass unit, atomic mass unit). Volatile substances were identified by comparison to the NIST MS Search 2.0 database in the mass spectrometer.

Abstract

The invention discloses a schizophyllum commune strain for generating a volatile bacteriostatic and nematocidal active component and application thereof, and belongs to the field of separation and application of the schizophyllum commune strain. The schizophyllum commune strain for generating the volatile bacteriostatic and nematocidal active component is separated from poplar leaves; the microbial preservation number of the strain is CGMCC No.12772. A GC-MS (Gas Chromatography-Mass Spectrometer) analysis demonstrates that volatile gas generated by the strain includes 16 effective components, wherein beta-bisabolol is the main component. Bacteriostatic and nematocidal test results show that the schizophyllum commune strain separated by the invention can effectively prevent and control various bacterial or fungal diseases which are harmful to plant growth, and the generated volatile product has remarkable inhibitory effects on worms and spawns of nematodes. The invention further discloses a biological fumigant prepared by using the schizophyllum commune strain and application thereof for preventing and controlling various diseases or parastic nematodes which are harmful to plant growth.

Description

technical field [0001] The present invention relates to Schizophyllum commune, especially a strain of Schizophyllum commune that produces volatile antibacterial and nematicide organic active ingredients. The invention belongs to the field of separation and application of schizophyllum, which controls the fungi, bacteria or nematodes which are harmful to the growth of plants. Background technique [0002] There are a large number of harmful organisms harmful to plant growth in the soil, such as harmful fungi, bacteria, parasitic nematodes, etc. The existence of these harmful organisms seriously endangers the growth of plants. [0003] The fumigation of chemical agents is effective in the control of plant diseases and insect pests. Among them, methyl bromide is a commonly used fumigant, which has high toxicity to harmful organisms, good penetration performance, and rapid diffusion. It has been widely used in the past. However, the large-scale use of methyl bromide will destro...
