Abstract

The invention provides a method for realizing high-accuracy fixed-point gene knockout in yeast. The method comprises the following steps: obtaining upstream and downstream sequences of a target knockout gene by a polymerase chain reaction or total-gene synthesis; connecting the upstream and downstream sequences in a direction opposite to the protogene, and inserting into a vector plasmid to construct an annular cloning vector; performing linear treatment on the annular cloning vector to obtain linear double-stranded DNA; performing 5'-to-3' exonuclease treatment on the linear double-stranded DNA to form linear 3' protruding long-cohesive end double-stranded DNA; and then converting yeast cells. In the invention, by use of the special exogenous DNA structure type, high-accuracy fixed-point gene knockout is realized in yeast. The accuracy of fixed-point gene knockout can reach 45-92%, the fixed-point gene knockout workload for yeast can be remarkably reduced, and the method is of relatively great application potential in yeast function gene study, gene engineering strain construction and the like.
