Problems solved by technology

For example: the medicinal materials are first degreased by petroleum ether or ether Soxhlet extraction, then heated and refluxed with methanol or 70% ethanol, and then extracted with ethyl acetate or n-butanol to obtain total flavonoids. Extraction and separation of total flavonoids from Suanzaoren not only has poor reproducibility, but also has many impurities in the extract and the flavonoid content is not high

Embodiment 1

[0029] A method for producing saponins and flavonoids by fermenting endophytic bacteria from wild spinous jujube, the steps are as follows: select uncultured Bacillus sp. extracted and purified from wild wild spiny jujube, GenBank accession number: KY393358; its 16S rDNA diagram is as follows image 3 shown;

[0030] (1) Activation of bacteria

[0031] The strains stored at 4°C were transferred from slant medium to solid medium, and cultured in a constant temperature incubator at 30°C for 12-20 hours. The composition of the medium is: 1% glucose, 0.25% yeast powder, 0.5% peptone, 0.06% NH 4 NO 3 , 0.5% KH 2 PO 4 , 0.01% MgSO 4 , 1.5% agar, and the rest is water. pH 7.0-7.2, sterilized at 115°C for 25 minutes.

[0032] (2) Preparation of seed solution

[0033] Take the above-mentioned activated strains and inoculate them into the seed culture medium (filling volume: 100mL / 500mL Erlenmeyer flask), shake the shaker at 180r / min, and incubate at 30°C for 12-20h. The compos...

Abstract

The invention discloses a method for producing saponins and flavonoids through wild jujube endophyte fermentation. An uncultured Bacillus sp. strain with the GenBank accession number of KY393358 is used, and the method for preparing the saponins and the flavonoids through the endophyte fermentation comprises the following steps: (1) activating a culture; (2) preparing a seed solution; (3) fermenting; and (4) refining the saponins and the flavonoids: centrifuging, filtering, performing alcohol precipitation, extracting, performing low-pressure concentration, performing chromatography, crystallizing and drying the saponin components and the flavonoid components to obtain the high-purity saponins and the high-purity flavonoids. The invention creates a novel way for large-scale production of the saponins and the flavonoids through the wild jujube endophyte fermentation; and by the method, with glucose as a raw material, the saponins and the flavonoids are prepared by an endophyte fermentation technology, a fermentation solution is simple in component, the products exist in the fermentation solution, few impurities are produced, and the products are easy to purify.

Description

technical field [0001] The invention relates to the technical field of wild jujube processing and production of saponins and flavonoids, in particular to a method for fermenting wild jujube endophytes to produce saponins and flavonoids. Background technique [0002] Wild jujube [Ziziphus jujuba Mill.var.spinosa (Bunge) Hu ex H.F.Chow.] is a kind of shrub or small tree belonging to Rhamnaceae Jujuba. It is widely distributed in the central and northern regions of my country. It has strong stress resistance. It is a wild medicinal plant that is resistant to drought, cold, and salt and alkali. It has high economic value and medicinal value. Use valuable wild fruit tree resources. It is recorded in the classic of traditional Chinese medicine "Shen Nong's Materia Medica", that jujube can "safety the five internal organs, lighten the body and prolong life", and the jujube seed is used for insomnia, palpitations, dreaminess, body deficiency and hyperhidrosis, and thirsty wounds. It ...
