Abstract

The method is to make human amniotic membrane into a suspension of human amniotic epithelial stem cells; place the cells in α‑MEM supplemented with endothelin 3, dexamethasone, stem cell factor, basic fibroblast growth factor, insulin transferrin and fetal bovine serum medium; at 37°C, 5% CO 2 Amniotic epithelial stem cells were cultured into an adherent monolayer in an incubator, and gradually induced to differentiate into cells expressing melanocyte marker genes tyrosinase-related protein 1 (trp1), 2 (trp2), microphthalmia-related transcription factor (mitf), tyrosine acidase (dtc) positive melanocytes.
