Embodiment

[0024] The tetanus immunoglobulin and the perfluorinated compound are connected to the carrier by a chemical connection method, and the chemical connection method is as follows:

[0025] 1) Pretreatment of agarose microspheres:

[0026] Wash the agarose microspheres with a large amount of distilled water to remove the organic solvent, and drain;

[0027] 2) Carrier activation:

[0028] Mix agarose microspheres, 1-4mol / NaOH solution, and epichlorohydrin at a volume ratio of 2:1:1, and react at 40°C for 1 hour; after the reaction, wash with a large amount of distilled water to obtain the activated modification agarose microspheres;

[0029] 3) Coupling of immunoglobulin and perfluoro-n-octylaniline on the carrier:

[0030] Dilute tetanus immunoglobulin to 150IU / mL with normal saline, and prepare tetanus immunoglobulin, perfluoro-n-octyl aniline and activated agarose microspheres according to the volume ratio of tetanus immunoglobulin:perfluoro-n-octyl Aniline (above 40°C): A...

Abstract

The invention discloses a tetanus exotoxin adsorbent. The tetanus exotoxin adsorbent is prepared from a carrier, and immune globulin and a perfluorinated compound which are immobilized on the surfaceof the carrier and are combined with tetanus exotoxin. A patient with tetanus is treated by utilizing an immunoadsorption method, and the human tetanus immune globulin is immobilized on a carrier microsphere; the free tetanus exotoxin in blood is efficiently cleaned by utilizing a principle of specifically combining the immune globulin and the exotoxin, and the perfluorinated compound is introduced so that the dissolved oxygen of an adsorbent is increased; the oxygen content in the blood is increased through extracorporeal blood circulation, and suffocation caused by laryngeal spasm on a patient is alleviated. The tetanus exotoxin adsorbent utilizes high-potency tetanus immune globulin; the immune globulin has strong specificity of neutralizing the tetanus exotoxin and has no anaphylacticreaction; the adsorbent has an additional oxygen aeration function so that the adsorbent is simpler to operate in clinical application process and the infection risks are smaller.

Description

technical field [0001] The invention relates to the field of medical biomaterials, in particular to a tetanus exotoxin adsorbent and its application. Background technique [0002] Tetanus is a specific infection often associated with trauma. It is an extremely serious disease with a high mortality rate. It is still a worldwide health problem. In developed countries, the main target of the disease is the elderly, and all of them are active immunized without booster immunization, while in developing countries, almost all people can get the disease, and tetanus is still an important cause of death for the poor. Due to the lack of effective treatment, the mortality rate of tetanus is 20% to 30%, and the mortality rate of severe tetanus patients is 70%. [0003] Tetanus is an acute toxic zoonotic disease caused by Clostridium tetani through deep wound infection. It is acute, fatal and preventable. Tetanus patients will have a series of complications: suffocation, lung infection...
