Embodiment 1

[0038] After being pre-cultured by starvation, Dunaliella salina is transported to the chemotaxis detection chamber through the two side branch pipelines of the "Ψ" channel with an external micropump, and seawater containing ammonium salt (NH4Cl) is transported through the central nutrient water sampling channel with an external micropump. Chemical effect detection room transportation. The fluid in the central nutrient flow at the micrometer scale exhibits laminar flow characteristics (Reynolds number Re is ~0.01), forming a laminar flow zone in the chemotaxis detection area. After the fluid is cut off, relying on diffusion, the nutrient line source concentration gradient that decreases from the center to the two sides of the channel is automatically generated in the chemotaxis channel. Dunaliella salina swims from both sides to the center after sensing the nutrients. When the ammonium salt concentration is 1μM-1mA, the chemotactic effect will occur within 1-2min, and the degre...

Embodiment 2

[0040] After being pre-cultured with starvation, Gucardia spp. is transported to the chemotaxis detection chamber through the two side branch pipelines of the "Ψ" channel by an external micropump. Transport to the chemotactic effect detection room. When the ammonium salt concentration is 0.2μM-1mA, the chemotactic effect will be produced within 1-2min, and the degree of chemotactic effect will be 5-6.

Embodiment 3

[0042] The water in the nutrient water reservoir and the injection channel is blocked by the diffusion point polyvinyl alcohol gel and the chemotactic effect observation chamber. 100μM nutrients can diffuse through the diffusion point to the chemotactic effect observation chamber to form a point source diffusion of nutrients gradient. Put the amino acid induced culture of Cryptomonas lanuginosa into the cell culture room. When the nutrients are Ala, Asp, Glu, Met and other amino acids, the degree of chemotactic effect is 3 to 4 within 2 to 3 minutes.

Abstract

The invention provides a microfluidic chemotactic chip, and further provides a water eutrophication indication method based on the microfluidic chemotactic chip. The microfluidic chemotactic chip is composed of three units of the nutrient water gradient forming unit, the cell sample-injecting unit and a chemotactic effect observation chamber. The nutrient water gradient forming unit can form one of a nutrient water linear source gradient or a nutrient water point source gradient. The cell sample-injecting unit is arranged at the corresponding position according to the gradient forming mode. The microfluidic chemotactic chip has the following beneficial effects that the microfluidic chemotactic chip can accurately simulate the nutrient water linear source gradient and the nutrient water point source gradient, and a friendly testing platform is provided for chemotaxis of microalgae and bacteria to nutrient substances; and the eutrophication degree of water is evaluated from the aspect ofthe biological phenomenon instead of exploration of a physicochemical environment, the more convenient and intuitive effects are achieved, and thus supplement is provided for an existing eutrophication evaluation method based on exploration of the physicochemical environment.

Description

Technical field [0001] The invention relates to the field of microfluidic chip technology and aquatic ecological environment evaluation, in particular to a water body eutrophication indicator method based on a microfluidic chemotactic chip. Background technique [0002] Various signs indicate that human activities are disrupting or changing the biogeochemical cycles of biogenic elements such as carbon, nitrogen, sulfur, and phosphorus on the earth, leading to a series of serious ecological effects and environmental degradation. Among them, the eutrophication of water bodies has become one of the main environmental problems that plague all countries in the world. Many researchers have conducted research on eutrophication evaluation and have proposed a variety of eutrophication evaluation models. Among them, the concentration of nutrients such as nitrogen and phosphorus in seawater is still the most important monitoring and evaluation indicator for seawater eutrophication. The mos...
