The preparation method comprises the following steps: selecting a lactic acid bacteria probiotic strain as a leavening agent, inoculating the leavening agent into sterilized rhizoma polygonati slurry, and fermenting to obtain the rhizoma polygonati fermentation product, wherein the leavening agent is prepared by mixing lactobacillus plantarum and lactobacillus brevis according to the bacterial suspension volume ratio of 1: (0-10), after the leavening agent is inoculated into the sterilized rhizoma polygonati slurry according to an inoculation amount of 1%-30%, and performing fermentation for 10-80 h at the temperature of 20-36.5 DEG C.
