AI Technical Summary

Problems solved by technology

At present, the widely used and relatively mature technical means in the production of Anthurium seedlings mainly adopts the adventitious bud generation method, which has the advantages of high multiplication coefficient, etc. However, the adventitious buds need to be transferred to the rooting medium for cultivation. Complete plants are obtained, and as the number of subcultures increases, the mutation rate of offspring also increases. The adventitious bud regeneration system has defects in genetic transformation, and it is easy to produce chimeric plants
[0005] The embryoid body generation technology can realize large-scale reproduction of plants, and can be applied to cell fusion, chromosomal variation and Agrobacterium-mediated genetic transformation. However, there are few reports on the embryoid body generation of Anthurium anthurium. Pinheiro, Martins, F.B., Cruz, A.C.F.D., Carvalho, A.C.P.P.D., Marília Contim Ventrella, Otoni, W.C.. (2013) Maturation of anthurium andraeanumcv. eidibel somatic embryos from nodal segments. In Vitro Cellular & Developmental, 39-Pla -312) Using stem segments with axillary buds as explants, embryogenic callus is induced on the medium supplemented with NAA, and then mature somatic embryos are obtained. However, the induction rate of embryogenic callus in this method lower

(Xin Weijie, Xu Bin, Wang Guangdong, Guo Weiming, Wen Fangde, Jin Jianping, (2006), Research on Somatic Embryogenesis and Plant Regeneration of Anthurium anthurium, Journal of Horticultural Science (06), 111-116.) by adding 2,4- D and KT induced somatic embryos from the leaves of Anthurium, but the number and degree of synchronization of somatic embryos were relatively low, and were greatly affected by genotype.

[0043] Examples and comparative examples

[0044] A method for inducing embryogenic callus generation and proliferation, and embryoid body generation and maturation at the root tip of 'Powder Champion' Anthurium anthurium, the cultivation steps are as follows:

[0045]1) Selection of explants: The explants came from the aseptic bottle seedlings of 'Pink Champion' Anthurium anthurium from the Tissue Culture Center of the Institute of Forestry and Fruit Trees, Shanghai Academy of Agricultural Sciences.

[0046] 2) Pretreatment of explants: Take out the well-rooted Anthurium tissue culture seedlings, cut off the root tip and place it in a sterile canned bottle, rinse it with sterile water for 3 times, and then place it on sterile filter paper to absorb excess water, cut the part of 0.5 cm from the root tip as an explant.

[0047] 3) Embryogenic callus induction: inoculate the c...

Abstract

The present invention discloses a tissue culture method for inducing anthurium andraeanum embryoids through root tips. The method comprises the following steps: anthurium andraeanum root tip selectionas explants, explant pretreatment, induction of embryogenic callus, proliferation culture, induction of embryoids, culturing and maturing, and finally regeneration of anthurium andraeanum plants. Anthurium andraeanum young tender root tips are subjected to isolated culture, callus production is stably induced, average induction rate reaches 80% or more, after propagation and culture, the embryoids can be induced, and each gram of the callus can be induced to produce average 40-50 embryoids. The method can enable the embryonic callus to be expanded on a large scale and to still have ability togenerate embryoids after several years of multiplication culture. The method establishes an efficient and stable anthurium andraeanum embryogenesis technology, can be applied to genetic transformation, cell mutation and fusion of anthurium andraeanum, can avoid formation of chimera plants, and is helpful for future genetic improvement research of the anthurium andraeanum.

Description

technical field [0001] The invention belongs to the field of induced regeneration of anthurium explants, and in particular relates to a tissue culture method for producing anthurium embryoids through root tip induction. Background technique [0002] Anthurium is a perennial herbaceous flower of the genus Anthurium of the family Araceae, native to the tropical rain forests of South America. There are many kinds of anthuriums, with various flower colors and patterns. The heart-shaped spathes are bright in color, beautiful in flower shape, and have a long-lasting flowering period. The leaves with velvet luster have a long growth cycle and have important ornamental value. They are usually used for fresh cut flowers and potted flowers. and flower beds. In the global flower market, sales of anthuriums are second only to orchids. [0003] The in vitro regenerative culture technology can quickly obtain a large number of healthy, neat and genetically consistent seedlings, which is ...
