Problems solved by technology

However, long-term single use of a certain chemical agent is likely to produce drug resistance and affect the control effect.
Due to the characteristics of citrus sand skin disease, it is necessary to apply pesticides multiple times within a year for prevention and control. Extensive use of chemical agents will cause pollution to the environment and potential harm to human health.

Embodiment 1

[0026] Example 1: Screening and Identification of Bacterial Strains

[0027] The screening method of the bacterium that the present invention has inhibitory action to citrus sand skin disease is as follows:

[0028] 1. Collection of citrus leaves

[0029] Healthy citrus leaves were collected from different orange orchards where citrus sand skin disease occurred in Mayang County, Huaihua City, Hunan Province, and brought back to the laboratory.

[0030] 2. Isolation of bacteria

[0031] Separation by dilution. Rinse the leaves with running water, soak them in 75% alcohol for 30 seconds, soak them in 1% sodium hypochlorite solution for 4 minutes, rinse them with sterile water for 5 times, and cut them into pieces about 1cm in size. 2 Place the small pieces in a sterilized mortar for grinding, dilute with sterile water, and prepare 10 -2 、10 -3 、10 -4 Dilution solution, draw 100 μL of the dilution solution and add dropwise to NA medium (beef extract 3g, agar 20g, sodium chl...

Embodiment 2

[0060] Embodiment 2: application embodiment

[0061] Carry out the field experiment of the fermented liquid of Bacillus subtilis (Bacillus subtilis) M-23 bacterial strain preventing and treating citrus sand skin disease, the test site is located in Huaihua City, Hunan Province Hongjiang City Shuangxi Citrus Horticultural Field, the orange orchard was built in 1986, the variety is Bingtang orange and citrus sand skin disease occurred more seriously. The field test was sprayed 5 times in mid-March 2019 when the spring shoots were 2-3cm long, in mid-April, when the flowers had faded to 2 / 3, when the fruit was the size of a broad bean, and when the fruit was the size of a table tennis ball. The control is carried out simultaneously, and the leaves are sprayed with an artificial knapsack electric sprayer, and each citrus tree is evenly sprayed with 3L. Set 1 control group and 2 treatment groups, treatment group 1: dilute Bacillus subtilis (Bacillus subtilis) M-23 fermented liquid ...

Abstract

The invention discloses a bacillus subtilis strain with an inhibiting effect on Diaporthe citri, a screening method and an optimal fermentation condition. The strain is preserved in the China GeneralMicrobiological Culture Collection Center (CGMCC); the preservation number of the strain is CGMCC No. 17205. The fermentation liquor of the strain can be used for safely, non-toxically and effectivelypreventing and treating the Diaporthe citri, and a field test shows that the prevention and treatment effect of the fermentation liquor diluted by 30 times on the Diaporthe citri is 79.09%; and the prevention and treatment effect of the 50-time diluted fermentation liquor on Diaporthe citri is 68.97%. Meanwhile, the strain has broad-spectrum antibacterial activity and also has a certain inhibiting effect on peanut sclerotium rolfsii, peanut crown rot, rice sheath blight disease and strawberry botrytis cinerea. At present, no report about the prevention and treatment of the Diaporthe citri bythe bacillus subtilis exists in China. Therefore, the antagonistic strain has a high application prospect.

Description

technical field [0001] The invention relates to a new bacterial strain with inhibitory effect on citrus psoriasis, a screening method thereof, and optimum fermentation condition optimization. Background technique [0002] The pathogenic form of citrus sand skin disease is Diaporth citri, isolated by Wolf et al. The asexual form of the pathogen is Phomopsis citri, isolated by Fawsett et al. Spore organs are formed under the epidermis, spherical, oval or irregular, with tumor-like orifices, conidia are colorless and transparent, and have two forms: α-type, spores are oval or spindle-shaped, single spores, general Contains 2 oil globules, sometimes multiple oil globules, easy to germinate; β-type, upright and slender or curved in a hook shape, without oil globules, this type of spores are not easy to germinate. Generally, α-type spores are mostly, and β-type spores are rare. The optimum growth temperature for pathogenic bacteria is around 25°C, higher than 35°C or lower than ...
