Embodiment 1

[0026] Example 1: Optimal Melatonin Application Concentration for Controlling Cucumber Green Mottle Mosaic Virus

1. Cultivation of Nicotiana benthamiana and cucumber

N. benthamiana and cucumber seeds were germinated at 25°C for 7 days in the dark, and seedlings of uniform size were selected and transferred to plastic pots (8 cm in diameter and 10 cm in height) containing a soil mixture containing vermiculite and pearl salt, Plant in a growth chamber at 22°C, 16 h / 8h, light / dark cycle.

[0027] 2. Friction inoculation with CGMMV

(1) Infiltrate CGMMV Agrobacterium on healthy 4-5 leaf stage N. benthamiana.

[0028] (2) After the onset of N. benthamiana, the diseased leaf tissue was taken and placed in a mortar, sprinkled with an appropriate amount of emery, and then ground.

[0029] (3) After the leaf tissue is ground, add an appropriate amount of sterile water, let it settle for 5 minutes, and absorb the supernatant.

Abstract

The invention belongs to the field of plant protection, and relates to a method for preventing and treating cucumber green mottle mosaic virus, in particular to application of melatonin to prevention and treatment of cucumber green mottle mosaic virus and a method for enhancing the prevention and treatment effect in cooperation with melatonin. The method comprises the implementation steps that a melatonin solution is irrigated to plant seedlings and roots or sprayed to leaf surfaces once a day, and treatment is conducted for three days in total. The invention further discloses a method for enhancing the prevention and treatment effect on the cucumber green mottle mosaic virus in cooperation with melatonin. The prevention and treatment effect of the melatonin on the cucumber green mottle mosaic virus is remarkably enhanced through the silent gene NbCRISP1. According to the application, the relationship between the non-toxic macromolecular melatonin and the CGMMV is taken as a research object, the effect of the melatonin in prevention and treatment of the CGMMV is defined, and the application of the melatonin in prevention and treatment of plant diseases is expanded.

Description

technical field [0001] The invention belongs to the field of plant protection and relates to a method for preventing and treating cucumber green mottle mosaic virus, in particular to the application of melatonin in preventing and controlling cucumber green mottle mosaic virus and a method for enhancing its control effect with melatonin. Background technique [0002] Cucumber green mottle mosaic virus (CGMMV) is an important imported and domestic quarantine virus in my country, which infects cucurbit crops and causes serious harm. After CGMMV infection, the leaves of the plants showed symptoms such as macula, mosaic leaves, chlorosis, and vesicles, and some may produce green tumor-like protrusions, which made the surface of the leaves uneven; the internal pulp often appeared discolored, fibrotic and rotten. In 2006, it was introduced into my country's Liaoning region. In recent years, it has shown a trend of expanding from local areas to the whole country, and spreading from ...
