The invention discloses a preparing method and an application of a sample for detecting cucumber green mottle mosaic viruses (CGMMV) carried by cucurbitaceae seeds. The preparing method includes the steps that starting from treating seed samples, shell-kernel separating treatment is carried out on the cucurbitaceae seeds, seed kernels with the low virus content are removed, seed shells with the high virus content are left to serve as to-be-tested samples, the seed samples obtained after treatment are sufficiently grinded through a full-automatic rapid sample grinding instrument, and the CGMMV in the large quantity of cucurbitaceae seeds can be rapidly detected through the sufficiently-grinded samples with a RT-PCR method and can also be rapidly detected through the sufficiently-grinded samples with a dot enzyme-linked immunosorbent assay (dot-ELISA) method. The preparing method and the application are suitable for virus detection of the common cucurbitaceae seeds such as calabash seeds, cucumber seeds, lagenaria sicerariae seeds, pumpkin seeds, long gourd seeds and watermelon seeds. A detecting method is high in sensitivity, good in specificity, short in consumed time and low in cost, technical supports are provided for large-scale detection and scientific prevention and control of the CGMMV carried by the cucurbitaceae seeds, and references are provided for detection of plant viruses transmitted by other seeds.

The invention discloses an immune colloidal gold test strip for quickly detecting a cucumber green mottle mosaic virus (CGMMV) and a preparation method of the immune colloidal gold test strip. The preparation method comprises the following steps: dripping a purified CGMMV polyclonal antibody solution on a detection line of a nitrocellulose membrane; dripping a goat anti-rabbit polyclonal antibody on a quality control line of the nitrocellulose membrane; sequentially attaching a sample pad made of water-absorbing filter paper, a CGMMV immune colloidal gold pad, the nitrocellulose membrane and water-absorbing paper to a polyvinyl chloride (PVC) adhesive plate with an adhesive sticker so as to prepare the test strip. The test strip can be used for detecting the CGMMV in a cucurbitaceous plant and is applicable to detection and diagnosis of a viral disease caused by the CGMMV. The method is simple and quick. The test strip is low in cost. A result can be obtained within 5 to 10 minutes and can be visually and directly observed by naked eyes. The test strip is applicable to quick and extensive screening and detection of diseases through basic agricultural departments at all levels, farmers and entry-exit inspection departments, facilitates timely and effective control and elimination of an epidemic situation, and has a wide application prospect.

The method comprises the implementation steps that a melatonin solution is irrigated to plant seedlings and roots or sprayed to leaf surfaces once a day, and treatment is conducted for three days in total.
