Example Embodiment

[0024] Example 1

[0025] Select mulberry leaves from Zhejiang, air-dry, pulverize into powder with a pulverizer, and pass through a 60-mesh sieve.

[0026] Weigh 50 g of mulberry leaf powder, add 1000 ml of 70% ethanol according to a material-to-liquid ratio of 1:20, and ultrasonically treat it for 10 min at 200 W power; control the extraction temperature to be 70 °C, and extract for 2 hours.

[0027] The extract was centrifuged at 4000 r / min for 10 minutes, and the supernatant was filtered overnight to obtain the mulberry leaf flavonoid filtrate. The content of flavonoids in mulberry leaves was 2.88% determined by spectrophotometry, and the calculated yield of flavonoids was 28.8 mg / g.

[0028] The flavonoid filtrate was concentrated under reduced pressure to obtain mulberry leaf flavonoid extract.

Example Embodiment

[0029] Example 2

[0030] Select mulberry leaves from Zhejiang, air-dry, pulverize into powder with a pulverizer, and pass through a 40-mesh sieve.

[0031] Weigh 50 g of mulberry leaf powder, add 2000 ml of 80% ethanol according to the ratio of material to liquid 1:40, and ultrasonically treat it for 20 min at 300 W power; control the extraction temperature to be 80 °C, and extract for 1 hour.

[0032] The extract was centrifuged at 4000 r / min for 10 minutes, and the supernatant was filtered overnight to obtain the mulberry leaf flavonoid filtrate. The content of flavonoids in mulberry leaves was 2.90% determined by spectrophotometry, and the calculated yield of flavonoids was 29 mg / g.

[0033] The flavonoid filtrate is dried in an oven at 50°C to 90°C to obtain the flavonoid extract.

Example Embodiment

[0034] Example 3

[0035] Select mulberry leaves from Zhejiang, air-dry, pulverize into powder with a pulverizer, and pass through a 60-mesh sieve.

[0036] 50 g of mulberry leaf powder was weighed, distilled water was added according to a material-to-liquid ratio of 1:40, and ultrasonically treated at 300 W for 10 min; the extraction temperature was controlled to be 80° C., and the extraction was performed for 1 hour.

[0037] The extract was centrifuged at 4000 r / min for 10 minutes, and the supernatant was filtered overnight to obtain mulberry leaf polysaccharide filtrate;

[0038] Add high-concentration ethanol to the polysaccharide filtrate to make the ethanol concentration in the final solution reach 70%, place at 4° C. for 18 hours, take the precipitate and dry, and the yield of mulberry leaf polysaccharide is 122.5 mg / g.

Abstract

The present invention discloses a method of extracting flavone and polyoses from mulberry leaves, including air drying, grinding and screening. Mulberry leaf powder is immersed in 60 to 80 percent ethanol so as to extract flavone or in water so as to extract polyoses; the ultrasonic treatment with 100 to 400W power lasts for 5 to 25 minutes; extraction temperature is controlled within 60 to 85 DEG C and the extraction lasts for 1 to 2 hours; the extracting solution is centrifugalized and filtered by supernatant fluid, thus obtaining the filtrate; the flavone filtrate is concentrated so as to obtain the mulberry leaf flavone extract; or ethanol is added into the polyoses filtrate so as to make the ethanol concentration up to 70 percent in the final solution; the filtrate is placed for 18 to 24 hours under 4 to 10 DEG C, then deposited and dried, thus obtaining the mulberry leaf polyoses. The flavone yield can reach more than 25mg/g and the polyoses yield 110mg/g. The present invention, with the advantages of good repeatability, high selection, short operating time, less solvent consumption, high yield of effective ingredients, no noise, no pollution and relatively good application prospective, is applicable to industrial production.

Description

technical field [0001] The invention relates to a method for extracting flavonoids and polysaccharides from mulberry leaves. Background technique [0002] Mulberry leaves account for about 64% of the output of the aboveground part of mulberry trees, and can be picked 3 to 6 times a year, with strong vitality. Mulberry leaves are rich in bioactive components such as flavonoids and polysaccharides. As one of the main functional components of mulberry leaves, flavonoids account for about 1.0-3.0% of the dry leaf weight. They are the ones with the highest content in all plant stems and leaves. They have anti-oxidation, lower blood pressure, prevent arteriosclerosis, Anti-aging and other effects. The rich polysaccharides in mulberry leaves have significant effects of lowering blood sugar and inhibiting blood lipid elevation, and have multiple regulatory functions in the treatment of type II diabetes. [0003] At present, the extraction of flavonoids and polysaccharides from mu...
