Problems solved by technology

However, using non-conductive materials to place between electrodes to prepare giant liposomes has many restrictions on non-conductive materials, and it is difficult to observe the giant liposomes formed on the non-conductive material layer; silicon is used as the base material to make array electrode structures , light cannot pass through the silicon substrate, and the preparation process cannot be observed under an inverted microscope. In addition, using silicon as the substrate material to make the surface array electrode structure requires professional processing, which is expensive and complicated
[0009] In summary, there are still some problems to be solved in the existing electro-preparation methods that use the chip structure to apply an electric field to prepare giant liposomes. The chips used in the early electro-preparation methods are simple and easy to process. However, the electro-preparation process It cannot be precisely controlled, and the improved chip proposed later can control the preparation process more precisely, but there are disadvantages such as difficult to observe, expensive, and complicated to operate.
In addition, all the chips that have been proposed do not include giant liposome sorting and collection parts. The sorting and collection of liposomes need to increase the post-processing steps, which increases the difficulty and investment, and affects the quality of giant liposomes. Sorting effect and final collection rate

Abstract

The invention discloses a device and a method for preparing and collecting lipidosome. The device comprises a signal generator and a preparation and collection chip, wherein the preparation and collection chip comprises a preparation chamber, a collection chamber and a waste fluid chamber; the preparation chamber comprises an electrode couple and a preparation cavity; the electrode couple is connected with the signal generator through electric conduction tapes; the collection chamber comprises an upper-layer collection cavity and a lower-layer collection cavity; a layer of microfiltration membrane is arranged between the two cavities; and the preparation chamber, the collection chamber and the waste fluid chamber are connected through microchannels. The device for preparing and collecting lipidosome mainly uses the method of powering up to prepare the lipidosome, and utilizes the microfiltration membrane to collect and process the lipidosome on the same chip. Compared with the prior art, the device and the method have the following technical effects: the large blank space lipidosome and the large lipidosome carried with drug, protein and particles can be effectively prepared; the preparation and collection of the lipidosome are integrated to reduce pilot process; and the required dose of reagents is less.

Description

technical field [0001] The invention relates to the field of liposome preparation, in particular to a device for preparing and collecting liposomes and a method for preparing and collecting liposomes using the device. Background technique [0002] Liposomes are closed vesicles in which a phospholipid bilayer membrane is assembled in an aqueous phase. Liposomes are amphipathic, biocompatible, and have a similar structure to cell membranes and organelle membranes, and are widely used in research fields such as beauty, food, medicine, and biochemistry. The particle size of liposomes ranges from 10 nm to 100 μm, and liposomes with a particle size larger than 5 μm are called giant liposomes. Giant liposomes can be used as carriers for drug delivery, and can also be used as microreactors to simulate cells or organelles, to study biochemical reactions in vivo, etc. In addition, giant liposomes can also be used to simulate cell membranes for cell membrane structure, function and R...
