[0023] In addition, in the preparation method of the fermentation broth of the microorganism provided in the embodiment of the present invention, it is preferable to activate the preserved microorganism before preparing the primary mother liquor. Compared with the microorganisms in preservation, the activated microorganisms have higher metabolic activity, and when they are used to prepare the first-grade mother liquor, their activity is higher, the adaptability is stronger and the reproduction speed is faster in the medium for the preparation of the first-grade mother liquor quick. It is beneficial to the preparation of the primary mother liquor.

[0024] The solid microbial agent of the microorganism provided in the embodiment of the present invention preferably uses corn starch as a carrier. As those skilled in the art can imagine, there are multiple acceptable carriers for preparing solid bacterial agents, such as cornstarch, potato starch, potato starch, etc.; wherein, co...

Embodiment 1

[0030] Embodiment one, the screening of this microorganism

[0031] The microorganisms provided by the embodiments of the present invention are obtained by screening by the following methods:

[0032] Chestnut nuts were collected from rose red chestnut orchard in Shengli Town, Luotian County, Huanggang City, Hubei Province, and stored at room temperature under controlled atmosphere. The seed coats and internal kernels of nuts without obvious chestnut rot were taken, and the endophytic bacteria were isolated and cultivated according to common methods in this field, and 5 kinds of endophytic bacteria were obtained, respectively numbered CMN1301, CMN1302, CMN1304, CMN1305, CMN1308. The pathogenic bacteria that cause solid rot in chestnut during storage: Phytophthora solani, Fusarium solani, Penicillium nigricans, Penicillium extensa, and Anthracnose were used as indicator strains, and plate confrontation culture was adopted according to the common methods in this field. Endophy...

Embodiment 2

[0039] Embodiment two, the identification of this microorganism.

[0040] (1) To analyze the morphological characteristics of this microorganism, the following steps can be followed:

[0041] Staining: Carry out Gram staining according to the routine method in this field, and observe under the microscope, the result is as follows figure 1 shown. As can be seen from the figure, the strain is red after Gram staining and is a Gram-positive bacterium.

[0042] Morphological characteristics: culture on LB medium at 37°C for 24 hours, observe the shape and color of the colony; take pictures of the bacteria, and observe the morphology of the bacteria with an optical microscope after staining ( figure 1 ). The results are described as follows: the colony is white, opaque, with small wrinkles. The bacteria are rod-shaped, contain a spore, and are motile.

[0043] (2) Analyze the physiological and biochemical characteristics of this microorganism, and operate according to the conve...

Abstract

The invention relates to the field of a microorganism, and particularly relates to a microorganism derived from Chinese chestnuts and an application thereof as well as a microbial agent preparation method. The microorganism derived from Chinese chestnuts is collected at the China Center for Type Culture Collection (CCTCC) on November 12, 2013 with the CCTCC No. M2013568, and the collection name of the microorganism is bacillus amyloliquefaciens subspecies CMN1308. The microorganism is an endophyte of Chinese chestnuts and has good effects in the terms of preventing and treating Chinese chestnut seed rot caused by at least one of cryponectria parasitica, fusarium solani, stachybotrys atra, penicillium expansum and colletotrichum, serving as a feed additive to promote the growth of animals and improve the immune ability and the like. Compared with the prior art using chemical pesticides for preventing and treating the seed rot, the microorganism does not cause the harm to human health and environmental pollution, and has important environmental protection significance.

Description

technical field [0001] The invention relates to the field of microorganisms, in particular to microorganisms derived from chestnut nuts, applications thereof and a preparation method of microbial agents. Background technique [0002] Chestnut (Castanea millissima. BL.) belongs to the genus Chestnut in the Fagaceae family. Known as the "King of Thousand Fruits", it is one of the important economic tree species in my country. Chestnut has a high water content, and it is easy to change color, taste and even rot during storage and transportation, which is academically called "real rot". The incidence of solid rot is very high, generally up to 20% to 30%, and up to 70% in severe cases, causing huge economic losses and seriously restricting the development of the chestnut industry. [0003] In the related art, chemical pesticides are mainly used to prevent and control the solid rot of chestnut. [0004] However, during the use of chemical pesticides, there are problems such as h...
