Abstract

The invention discloses a chromatographic method for determining the content of EPA, DHA and total w-3 acids in fish oil. The method comprises the following steps: 1, esterifying; 2, preparing a sample solution; 3, preparing a standard substance solution; 4, setting chromatographic conditions; 5, sequentially introducing samples; and 6, comparing the relation between the retention time and the peak area of a sample to be determined with that of a standard substance to determine the content in the sample. In the w-3 fatty acid-containing fish oil sample treatment process, unsaturated fatty acids is saponified, saponified unsaturated fatty acids and a methanol solution of boron trifluoride directly undergo an ester formation reaction, and unsaturated fatty acid esters are directly extracted by a BHT-containing isooctane solution to obtain a solution used as the sample to be determined. The method adopting the appropriate ester formation process and the sample post-treatment process has the advantages of short test operation cycle, effective avoiding of volatilization and other problems in the operation, and accurate and reliable determination result, and is especially suitable for industrial production and other occasions needing large scale determination.
