Embodiment Construction

[0030] The following describes the present invention in detail with reference to the drawings and specific embodiments.

[0031] 1. Screening method of CTL epitope peptide on Hantaan virus envelope glycoprotein

[0032] Step1: Forecast

[0033] The amino acid sequence of the glycoprotein of HTNV 76-118 strain was obtained from the NCBI database, and these amino acid sequences were input into the epitope prediction software IMMUNE EPITOPE DATABASE AND ANALYSIS RESOURCE using bioinformatics technology to predict the CTL epitope peptide composed of 8 amino acids, H -2b limit.

[0034] The prediction software predicts a total of 15 epitope peptides on HTNV GP. The amino acids of these 15 epitope peptides are:

[0035] (1) GP aa39~46: SVIGYVEL Serine-Valine-Isoleucine-Glycine-Tyrosine-Valine-Glutamic acid-Leucine;

[0036] (2) GP aa44~51: VELPPVPL valine-glutamic acid-leucine-proline-proline-valine-proline-leucine;

[0037] (3) GP aa64~71: SMDNHQSL serine-methionine-aspartic acid-asparagine-g...

Abstract

The invention discloses CTL epitope peptide on HTNV GP as well as a screening method and application thereof. The epitope peptide is composed of 8 amino acids: isoleucine-threonine-serine-leucine-phenylalanine-serine-leucine-leucine (ITSLFSLL), and the sequence position is GPaa 456-463; the screening method of the epitope peptide comprises the following steps of: prediction, synthesis and detection; and according to research, the epitope peptide can be applied to the stimulation of body cellullar immunologic response. The invention has the following beneficial effects: through the screening and killing ability detection of CTL epitope peptide on HTNV GP and in combination with abundant neutralizing epitopes of GP, the body can be stimulated to generate relatively strong neutralizing antibodies, the development of a novel multifunctional efficient peptide-fragment vaccine with neutralizing epitope and CTL epitope is facilitated, and the invention is of important significance to the research of a HTNV novel genetic engineering vaccine.

Description

Technical field [0001] The present invention relates to a CTL epitope peptide on Hantaan virus envelope glycoprotein (HTNV GP for short), a method for screening the epitope peptide, and the application of the epitope peptide, and belongs to the technical field of microbial immunity. Background technique [0002] Hantaan virus (Hantaan virus, HTNV) belongs to Bunyaviridae and Hantavirus. In 1976, Korean scholar Lee Ho-Wang successfully isolated the virus from the lung tissue of Apodemus agrarianus in the Hantan River Basin of the country's epidemic area. The host of the virus is rodents such as rodents, and it is mainly transmitted to humans through the respiratory tract, digestive tract and broken skin through the pollution of animal excrement, causing Hemorrhagic fever with renal syndrome (HFRS), mainly clinical Manifested with fever, bleeding and acute renal damage. my country is the country with the most severe HFRS epidemic in the world. It is characterized by a wide range ...
