Problems solved by technology

This is not only time-consuming and laborious (from the process of milk sample collection-stretch separation-enrichment culture-morphological observation-biochemical identification-drug susceptibility test needs at least 6-7 days), and because the occurrence of cow mastitis is often caused by many pathogenic bacteria, and conventional bacterial isolation procedures cannot cover all pathogenic bacteria, so that the antibiotics screened by the drug susceptibility test still have limitations in treatment
At the same time, due to the selection of different culture media for different bacteria, although specific bacteria can be obtained, more materials are consumed, resulting in increased costs for diagnosis and treatment

Embodiment 1

[0044] The growth curve determination of embodiment 1 dairy cow mastitis-associated bacteria

[0045] 1.1 Dip Staphylococcus aureus, Salmonella bovis, Escherichia coli bovis, Streptococcus uberis, Streptococcus dysgalactiae, Streptococcus agalactiae, Staphylococcus epidermidis, Cryptobacterium bovis, Bovis multocida with a 1μl inoculation loop. Bacillus bovis, Bacillus bovis and other 10 strains of bacteria liquid, streak pure culture on TSA+5% FBS medium.

[0046] 1.2 Pick a single colony and inoculate it into the common liquid enrichment medium for the main pathogenic bacteria of dairy cow mastitis, and then shake the bacteria overnight at 37°C and 200rpm.

[0047] 1.3 Repeat the above steps 2 times to obtain stable growing colonies

[0048] 1.4 Pick a single colony of the above strains and inoculate them into the common liquid enrichment medium for main pathogenic bacteria of cow mastitis, shake the bacteria at 200 rpm for 12 hours at 37°C.

Embodiment 2

[0054] Example 2 Drug Sensitivity Test of Bacteria Related to Dairy Cow Mastitis

[0055] 2.1 Pipette 100 μL of the 9 kinds of bacterial cultures and their mixed cultures and their bacterial mixtures in Example 1 respectively (add the same amount of general medium to make OD600=0.5) and smear on the solid medium plate of cow mastitis universal drug susceptibility test , after a little drying, stick the drug-sensitive paper to the surface of the agar with sterilized tweezers. Each plate is pasted with 7 pieces of paper, that is, one piece in the center and 6 pieces in the equidistant range around it. 3 plates for each sample, a total of 21 kinds of antibacterial drugs. In order to make the drug-sensitive sheet stick closely to the medium, gently press the drug-sensitive sheet with tweezers. Put the drug-sensitive tablet in a 37°C incubator overnight, observe and measure the diameter of the inhibition zone.

Abstract

The invention discloses a method for rapidly screening a sensitive antibiotic for treating cow mastitis. The method comprises the steps of acquiring a milk sample, implementing enrichment culture and drug sensitive testing. Two culture mediums with main pathogenic bacteria of the cow mastitis are prepared, a bacteria-growth curve test proves that the growth speeds of 10 main pathogenic bacteria related to the cow mastitis have no obvious difference, and then the method for rapidly screening the sensitive antibiotic for treating the cow mastitis is established on the culture medium. Compared with a conventional method, the time is shortened by 2-3 days, the workloads of separating the bacteria and implementing a drug sensitive test on the bacteria one by one are reduced, and the test materials are reduced. More importantly, the method enables all pathogenic bacteria to grow together at the same time, the screened sensitive antibiotic has an effect of inhibiting the bacteria causing the cow mastitis, and a better curative effect is provided clinically.

Description

technical field [0001] The invention relates to the technical field of prevention and treatment of cow mastitis, in particular to a method for rapidly screening sensitive antibiotics for treating cow mastitis. Background technique [0002] Cow mastitis is one of the main diseases affecting the development of dairy industry. According to the statistics of the World Dairy Cattle Association, about 1 / 3 of the dairy cows in the world suffer from various types of mastitis, and the economic loss caused by mastitis reaches 35 billion US dollars every year. According to reports, in the United States, the annual economic loss due to mastitis reaches 2 billion US dollars, and in New Zealand, the annual economic loss due to mastitis exceeds 300 million New Zealand dollars. Recent studies have found that the average incidence of mastitis in foreign dairy cows is 25% to 60%, and the economic loss caused by mastitis per cow in different countries is 61-93 pounds per year. The average pr...
