According to the invention, a TaqMan probe multicolor fluorescent PCR technology is adopted, a universal primer for enterovirus is designed in a relatively conserved sequence of viral sequences, and a universal type of enterovirus, enterovirus 71 and coxsackievirus A16 are detected byspecific probes. According to the kit, the enterovirus can be detected only by virtue of the detection of different fluorescent labeled probes in the detection process without melting curve analysis after the fluorescence PCR, and the enterovirus 71 can be distinguished from the coxsackievirus A16.
