[0052] The present invention also provides a method for preparing a protective repairing hydrogel for damaged parts of the skin and mucous membranes, comprising the following steps:

[0053] Step 1: dissolving the polymer matrix in triple distilled water to obtain an aqueous solution of the polymer matrix with a mass volume ratio of 0.5% to 5%;

[0054] Step 2: Add 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride and N-hydroxysuccinimide respectively to the polymer matrix aqueous solution, and react for 30-40 minutes ;

[0055] Step 3: continue to add 5'-hydroxydopamine, and adjust the pH to 4-5, and react overnight;

[0056] Step 4: dialyzing the reaction product obtained in the step 3 in an acidic phosphate buffered saline solution, the pH of the acidic phosphate buffered saline solution is 4-5;

[0057] Step 5: Add tissue repair-promoting drugs and anti-infective drugs to the dialysis product, mix evenly, and freeze-dry to make powder;

Abstract

The invention provides a protective repair hydrogel for damaged parts of skin and mucous membranes. The protective repair hydrogel comprises the following components in percentages by weight: 0.5% to10% of a polymer matrix, 0.001% to 0.1% of 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride, 0.001% to 0.1% of N-hydroxysuccinimide, 0.001% to 0.1% of 5'-hydroxydopamine, 0.01% to 5% of a drug for promoting tissue repair, 0.5% to 5% of an anti-infective drug and 80% to 98% of threefold-distilled water. The invention also provides a preparation method of the protective repair hydrogel for damaged parts of skin and mucous membranes, the weight percentages of 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride, N-hydroxysuccinimide and 5'-hydroxydopamine are optimized, and a layer of protective film can be formed by oxidative crosslinking when the surface of a wound is covered with the protective repair hydrogel, so that the failure of the drug for promoting tissue repair and the anti-infective drug is avoided, and the repair of the wound by the tissue repair drug and the anti-infective drug is also facilitated.

Description

technical field [0001] The invention relates to the technical field of biomaterials, more specifically, the invention relates to a protective repairing hydrogel for skin and mucous membrane damage and a preparation method thereof. Background technique [0002] In daily life, contusions, abrasions, scratches, cuts and other skin injuries often occur. In order to make the wound repair faster, repair hydrogel is often applied to the wound. [0003] Chinese invention patent 201710752030.1 discloses a hydrogel for repairing tissue damage and its preparation method. PGE2 powder is dissolved in phosphate buffer to make a PGE2 solution, which is added dropwise to the hydrogel and stored at 2-8°C or Freeze at -18--22°C, and the hydrogel material is at least one of chitosan, collagen, hyaluronic acid and fibrin. The hydrogel can prolong the release time of PGE2 to a certain extent, which can be directly used in the treatment of tissue damage and promote the recovery of damaged skin ...
