Abstract

The invention provides a digital chip and method for nucleic acid amplification. The digital chip for nucleic acid amplification includes a digital detection array reaction chamber, a main sample inlet and a main sample outlet, wherein the digital detection array reaction chamber includes multiple main channels and multiple micro-units, each micro-unit communicates with the main sample inlet and the main sample outlet through one main channel, and each micro-unit includes a micro-unit reaction chamber; and the inlet and outlet of each micro-unit reaction chamber separately communicate with thecorresponding main channel, and the width of the section, after the joint of the inlet of each micro-unit reaction chamber with the corresponding main channel, of the corresponding main channel is reduced, so that when a water phase in the corresponding main channel passes through the section, the water phase is introduced into each micro-unit reaction chamber, and the flow direction of an oil phase is unchanged. The fluid behavior of a liquid in the hydrophobic channels is controlled through an oil-water interfacial tension, therefore, a PCR reagent is stored in the designed micro-reaction chamber, and is sealed with oil, and the digital chip can be applied to nucleic acid amplification detecting experiments efficiently and conveniently; and the problems of a complex chip technology, high requirements of a manufacture process and inconvenient operation can be solved in the field of dPCR.
