Problems solved by technology

[0010] The inventor previously studied the effect of cannamycin on anti-inflammatory drugs and anti-coagulant drugs (see Chinese patent CN106377525A application of cannamycin in the preparation of anti-coagulant drugs, CN106176719A application of cannamycin in the preparation of anti-inflammatory application in medicines), and at the same time, a secondary metabolite (cannamycin) with certain activity has been obtained from the fungus (chaetomium attenuosa) collected from the soil of the Yellow River Delta (see Chinese patent CN106399118A, a kind of chaetomium attenuata and its application in the preparation of cannamycin), but its antitumor properties are not clear

Embodiment 1

[0049] Example 1 Antitumor activity of colamycin CA-1 and its effect on tumor cell proliferation and apoptosis

[0050] 1.1 Experimental method

[0051] 1.1.1 Cell Culture

[0052] Human colon cancer cells SW620 and human hepatoma cells HepG2 were cultured in DMEM medium (containing 1% penicillin-streptomycin double antibody, 10% FBS (fetal bovine serum)) at 37°C, 5% CO 2 , in a cell incubator with saturated humidity;

[0053] Human lung cancer cells A549, human lung cancer cells NCI-1975, and human lung cancer cells NCI-1650 were cultured in RPMI-1640 medium (containing 1% penicillin double antibody, 10% FBS (fetal bovine serum)) at 37°C, 5 %CO 2 , in a humidified cell incubator.

[0054] 1.1.2 MTT assay to analyze the inhibitory effect of colamycin CA-1 on the proliferation of various tumor cells

[0055] The cultured human colon cancer cells SW620, human liver cancer cells HepG2, human lung cancer cells A549, human lung cancer cells NCI-1975, and human lung cancer cell...

Embodiment 2

[0070] Example 2 The effect of co-administration of colamycin CA-1 and cisplatin on the proliferation of colon cancer cells

[0071] 2.1 Experimental method

[0072] 2.1.1 See 1.1 in Example 1 for cell culture.

[0073] 2.1.2 Investigate the effects of single use of clokamycin CA-1 or cisplatin, and their combination on the proliferation of colon cancer SW620 cells

[0074] During the experiment, the cells were treated with an equal concentration gradient of colamycin CA-1 and cisplatin alone or in combination, and the cell proliferation was analyzed by the MTT method. The specific steps are as follows:

[0075] Take the human colon cancer cells SW620 cultured in vitro, centrifuge, discard the supernatant, collect the cells, suspend, count the number of cells with a hemocytometer, 1 × 10 per well 4 The cells were seeded in a 96-well plate, allowed to grow adherently for 24 hours, and then added to 100 μL of medium containing different concentrations of CA-1 or / and cisplatin fo...

Abstract

The invention belongs to the technical field of medicinal chemistry, and particularly relates to application of cochliodinol in preparation of antitumor drugs. According to the present invention, the cochliodinol is singly used or the cochliodinol and the cis-platinum (or the oxaliplatin) are combined to act on different tumor cells so as to provide different inhibition effects, such that the broad anti-tumor effect is provided, and the inhibition effect on the human colon cancer cell SW620 is strongest. Experimental results show that cochliodinol can effectively reduce expression of p-MEK protein, p-ERK protein and apoptosis protein bcl-2, so that cochliodinol can act on tumor cell apoptosis and cycle processes to play an anti-tumor role. Experimental results show that cochliodinol and cis-platinum are combined to have a relatively strong inhibition effect on proliferation of human colon cancer cells SW620, and a new method and strategy are provided for colon cancer treatment by combining cochliodinol and cis-platinum.

Description

technical field [0001] The invention belongs to the technical field of medicinal chemistry, and in particular relates to the application of colamycin in the preparation of antitumor drugs. Background technique [0002] Colon cancer is a common malignant tumor of the digestive tract, which occurs mostly at the junction of the rectum and the sigmoid colon, and ranks third in the incidence of gastrointestinal tumors. Colon cancer is mainly adenocarcinoma, mucinous adenocarcinoma, undifferentiated carcinoma, and the general shape is polypoid and ulcerative. Colon cancer can develop in a circle along the intestinal wall, spread up and down along the longitudinal diameter of the intestinal canal, or infiltrate deep into the intestinal wall. In addition to lymphatic vessels, blood flow metastasis and local invasion, it can also be implanted in the abdominal cavity or spread along the suture and incision surface. . [0003] In the prior art, conventional colon cancer treatment met...
