Embodiment Construction

[0023] Below by example the present invention will be further described:

[0024] Isolation, screening and identification of strains

[0025] The strain isolation and screening method of the present invention is as follows: place the Porphyra algae collected from seawater in a sterilized mortar, add an appropriate amount of sterilized seawater, stir and grind, and spread the serial concentration of the slurry on a 2216E seawater agar plate, After culturing at 25° C. for a certain period of time, the colonies with transparent circles or depressions around them were picked for further isolation and purification of strains.

[0026] The whole-cell fatty acid composition analysis of the strains uses the Microbial Identification System system, and the utilization of various carbon sources by the strains uses the Biolog system, and the colonies directly picked from the 2216E plate are used.

[0027] The primers used for 16S rDNA sequence analysis were universal primers 27F (5′-AGAG...

Abstract

The invention discloses a novel flavobacterium strain, a cultivation method for the strain and application of the strain in the aspect of producing agarase. The provided strain which can be used for producing the agarase is flavobacterium sp. CGMCC NO.2342, and is specially characterized in that the strain can generate the agarase without agar, and 0.05 to 0.25 percent of agar powder or a certain carbon source can be added in a liquid culture medium. The provided method for producing the agarase cultivates the flavobacterium sp. in a fermentation culture medium; and the crude enzyme liquid of the obtained agarase can be continuously concentrated and purified, and also can be directly used for degrading the agar to produce agar oligosaccharide. The agarase produced by the strain has high activity, produces various agarase, and can be expected to form various series of agar oligosaccharide.

Description

technical field [0001] The present invention relates to a new microorganism, in particular to a bacterium of the genus Flavobacterium (Flavobacterium sp.), and also relates to the application of the bacterial strain in producing agarase. Background technique [0002] Agar gum is a water-soluble polysaccharide extracted from red algae such as Asparagus (asparagus) and Geliflower, and is one of the world's three major seaweed gums. It is widely used in biochemical, clinical, pharmaceutical, food and other fields. Because agar is not easy to be degraded by most microorganisms, it is also widely used as a laboratory medium support carrier. [0003] Agarase is a class of enzymes that can degrade agar polysaccharides. Agarase is mainly divided into two categories: a-agarase and β-agarase according to the difference in the cleavage of glycosidic bonds. α-agarase cleaves the a-1,3 glucosidic bond of agarose, and β-agarase cleaves the β-1,4 glycosidic bond of agarose. Agarase is ma...
