According to the method, tissue like a stem of Caesalpinia cucullata Roxb. is cultured on an N6 culture medium, and a rhizome with one or two buds is exactly taken as an explant, then disinfected and inoculated into a culture medium consisting of N6, 0.5 mg / L of 2,4-D, 1.4 mg / L of 6-BA and 0.4 mg / L of IBA for culture; the bud germinates in about 6 to 8 days after inoculation, and a young seedling with leaves is formed in about 10 days; the young seedling is cut down and inoculated into a culture medium consisting of N6, 0.1 mg / L of 2,4-D, 1.8 mg / L of 6-BA and 0.4 mg / L of IBA so as to maintain high-speed progressive increase, and a propagation coefficient is more than 5; a culture medium consisting of N6, 0.5 mg / L of 6-BA and 1.5 mg / L of NAA is selected as a rooting medium, a rooting rate reaches more than 90%, and a complete plant is formed in only about 25 days; and a test-tube plantlet is transplanted into sandy soil, attention is paid to water retention and moisture retention, a transplanting survival rate reaches more than 90% in one month after transplanting, and a high frequency stable regeneration system is established.
