Embodiment 1

[0027] 1. Plant materials and multiple stress treatments

[0028] The 1-year-old Ginkgo seedlings were cultivated in the greenhouse of Nanjing Forestry University, and the roots, stems, leaves, petioles, and kernels and buds of 25-year-old Ginkgo trees in the school were used as materials for gene cloning and expression analysis. In addition, Ginkgo seedlings were treated with drought, salt, darkness, salicylic acid (SA) and methyl jasmonate (MJ). Three biological replicate experiments were performed at each sampling time under different stress conditions. Seedlings were treated with 20% (w / v) PEG (MW 6000) and salinity (200 mmol / L NaCl). Dark treatment is to place the plants in a dark room. The exogenous hormone treatment was to spray 100μM / L SA and MJ on the leaves of Ginkgo seedlings respectively. Other conditions of the treated seedlings were similar to those of the control seedlings. Treated and control seedling leaves were collected at 0, 3, 6, 12, 24 and 48 h after ...

Abstract

The invention discloses a gingko gene GbBBX25 and an expression protein and application thereof. The base sequence of the gingko gene GbBBX25 is shown in SEQ ID NO.1. The gene GbBBX25 is separated from gingko, codes 819bp open reading frames (ORFs) of 273 amino acids and contains two b-box structural domains but contains no CCT structural domains. The GbBBX25 is located in cell nucleuses and has important nuclear localization characteristics of transcription factors. The expression quantity of GbBBX25 transcripts is highest mainly in leaves, and the inductive effect under salt stress is remarkable. The GbBBX25 is over expression in Populus davidiana*P.bolleana, it is proved that under salt stress, the soluble sugar content, the total protein content and the peroxidase (POD) activity of thetransgenic poplar are higher than those of non-genetically modified organism poplar, and it is shown that the GbBBX25 can improve the salt resistance by improving the efficiency of an antioxidant system. Overexpression of the GbBBX25 in the transgenic poplar can improve the salt resistance of plants, improvement on the expression level of the gene GbBBX25 can be used in breeding for abiotic stress, and the gingko gene GbBBX25 is high in practicability.

Description

technical field [0001] The invention belongs to the technical field of genetic engineering, and in particular relates to a ginkgo GbBBX25 gene, its expressed protein and its application. Background technique [0002] Plants have evolved a complex network that responds to unstable abiotic and biotic conditions with highly complex mechanisms. Abiotic stresses such as drought, high salinity, and low temperature are important factors affecting plant growth and development. In breeding work, it is not only necessary to breed high-yielding, high-quality varieties, but also to breed high-resistant varieties that can adapt to various adversities. Zinc finger proteins play an important role in regulating plant growth and development and coping with stress. B-box (BBX) proteins are zinc finger transcription factors or transcriptional regulators that contain one or two B-boxes. In 1995, the first B-box protein (CO) was discovered in Arabidopsis. In 2009, 32 proteins with N-terminal ...
