[0103] Example 2 - Evaluation of Biosensor System Precision

[0104] Protocol: The precision of the biosensor system was assessed over at least 20 days (not necessarily consecutive working days), with two runs per test day, with at least two hours between each run, for each sample per run Repeat twice. Based on this design, a total of 80 measurements were obtained per sample (20 or more days x 2 runs per day x 2 results per run). Use a single instrument at a single location throughout the testing process.

[0105] On the morning of Day 1, the first run was performed using freshly prepared commercial BNP-free plasma samples spiked with 500 pg / ml BNP. In the afternoon of Day 1, a second run was performed using another freshly prepared commercial BNP-free plasma spiked with 500 pg / ml BNP. Measurements were repeated twice in each run. This was repeated for another 19 days. Each replicate of EIS data is recorded in a nested data recording table.

Abstract

A biosensor useful for the detection of a target analyte in a fluid biological sample is provided. The biosensor comprises a first sensing layer comprising at least a pair of metal electrodes on an inert substrate, wherein the electrodes are linked by an electrically conductive bridge having immobilized on the surface thereof analyte-reactive compounds, wherein the electrically conductive electrode bridge comprises a material that responds to binding of target analyte to the analyte-reactive compound with an electrochemical or electrical change; and, optionally, a second layer comprising one or more sample chambers for placement on the first sensing layer, wherein the sample chambers are adapted to receive a fluid sample and direct the sample to the bridge. The biosensor may be incorporated in a point of care device additionally comprising a detection device that forms an electrical circuit with the biosensor, provides an electrical signal over a range of frequencies and is adapted todetect an electrochemical or electrical change at the electrode bridge in the presence of target analyte using electrochemical impedance spectroscopy on application of the electrical signal.

Description

technical field [0001] The present invention relates generally to sensing systems and methods, and in particular, to novel biosensors, devices incorporating biosensors, and methods for detecting analytes in samples. Background technique [0002] Heart failure (HF) occurs when the heart can no longer pump blood adequately to meet the body's needs. HF is a global pandemic affecting at least 26 million people worldwide with increasing prevalence. The cost of HF to the economy is significant and will expand dramatically as the population ages. Despite significant advances in treatment and prevention, patients continue to suffer from poor quality of life and high mortality and morbidity. Accurate and early diagnosis is crucial to improve the outcome of HF patients. However, HF diagnosis can be challenging because HF symptoms such as shortness of breath (dyspnea) or fatigue are subjective and nonspecific. For example, dyspnea is seen in individuals who are obese or have COPD b...
