Embodiment Construction

[0028]The invention provides a novel tumor imaging and therapy probe FSH-Rh760 targeting FSH receptor, which is formed by coupling FSH polypeptide and near-infrared fluorescent dye Rh760.

[0029] The specific embodiment of preparing the above-mentioned near-infrared fluorescent probe is as follows:

[0030] (1) 0.2 mmol of compound Rh760, 100 mg (0.25 mmol) of Carter condensation reagent, 150 mg (1 mmol) of piperazine and 10 mL of dichloromethane solution were added to a 50 mL round-bottomed flask, and the reaction was stirred at room temperature for 2 h. After completion, the solvent was distilled off under reduced pressure, and the compound Rh760-Paz (105 mg) was obtained by purification and separation by column chromatography using dichloromethane and methanol as eluents. Yield: 80%. 1 H NMR (400 MHz, DMSO-d6, δ) 8.99 (bs, 1H), 7.62 (m, 2H), 7.53 (d, J = 8.4 Hz, 1H), 7.35 (d, J = 8.4 Hz, 1H), 6.76 (dd, J =9.2 Hz, J = 2.4 Hz, 1H), 6.71 (d, J = 9.2 Hz, 1H), 6.62 (...

Abstract

In order to solve the problem of accurate identification of lesions during surgery, on the basis of the advantages of near-infrared fluorescent dyes for imaging, its specific imaging is increased. The invention provides a tumor imaging and treatment probe targeting follicle stimulating hormone receptor and a preparation method thereof. In the present invention, the follicle stimulating hormone polypeptide (LYTRDLVYKDPARPKIQKTCTF) is coupled with the near-infrared fluorescent dye Rh760 to form the follicle stimulating hormone receptor-targeted tumor imaging and therapy probe FSH-Rh760. The advantage is that the present invention utilizes FSHR to be highly expressed in ovarian cancer , based on the principle of specific binding of FSH polypeptides to FSHR, targeting ovarian cancer cells. Utilizing the advantages of deeper penetration depth and lower background signal of the near-infrared fluorescent dye Rh760, it has a good application prospect in fluorescence imaging and fluorescence-guided surgery. The targeting probe FSH-Rh760 of the present invention can be used for photothermal therapy of ovarian cancer, as a supplement to surgical treatment, to improve the prognosis of ovarian cancer.

Description

technical field [0001] The present invention relates to a tumor imaging and treatment probe targeted by follicle stimulating hormone receptor and its preparation method and application. Background technique [0002] The mortality rate of ovarian cancer ranks first among malignant tumors of female reproductive system. Eighty-seven percent of cases were detected at an advanced stage (stages III and IV), and the 10-year survival rate was only 15%. Debulking surgery combined with chemotherapy is the main treatment modality for advanced ovarian cancer. The thoroughness of surgery is closely related to the prognosis of ovarian cancer. In order to achieve complete resection, accurate identification of the lesion during surgery is the key. Existing imaging examinations such as ultrasound, X-ray, CT, MRI, etc. are non-specific imaging of tumors, and are difficult to be used for intraoperative real-time imaging. Fluorescence-guided surgery uses tumor-targeted imaging probes that s...
