Embodiment 1

[0027] Example 1 Preparation of Malachite Green Molecularly Imprinted Polymer Microspheres

[0028] Add 2mmol of malachite green, 8mmol of acrylamide and 8mmol of methacrylic acid into a 250mL conical flask, and then add 15mL of petroleum ether. Measure 150 mL of distilled water, add 4.5 mL of Tween-80 (according to 3% of the volume of distilled water), heat to completely dissolve, and cool to room temperature. Distilled water was poured into a conical flask, and a crosslinking agent ethylene glycol dimethacrylate (40 mmol, 7.56 mL) and an initiator azobutadiisodinitrile (200 mg) were added. Ultrasonic degassing for 5 minutes, emulsification with a dispersing emulsifier for 3 minutes, and polymerization in a water bath at 65°C for 24 hours. The obtained polymer was washed with distilled water until clear, and then Soxhlet extraction was performed with 80% acetonitrile-ammonium acetate buffer as the washing solution. Finally, wash with 80% acetonitrile-ammonium acetate buffer...

[0029] Example 2 Preparation of Malachite Green Imprinted Molecular Polymer Microspheres

[0030] Add 2mmol malachite green, 4mmol acrylamide and 4mmol 4-vinylpyridine to a 250mL Erlenmeyer flask, then add 15mL chloroform, measure 150mL distilled water, add 7.5mL Tween-80 (according to 3% of distilled water volume), heat Make it completely dissolved and cool to room temperature. Distilled water was poured into a conical flask, and a crosslinking agent ethylene glycol dimethacrylate (40 mmol, 7.56 mL) and an initiator azobutadiisodinitrile (200 mg) were added. Ultrasonic degassing for 10 minutes, emulsification by a dispersing emulsifier for 5 minutes, and polymerization in a water bath at 70°C for 4 hours. The obtained polymer was washed with distilled water until clear, and then Soxhlet extraction was performed with 80% acetonitrile-ammonium acetate buffer as the washing solution. Finally, the solid-phase extraction device was washed with 60% acetonitrile-ammonium acetate b...

[0031] Example 3 Preparation of Malachite Green Imprinted Molecular Polymer Microspheres

[0032] Add 2mmol malachite green, 4mmol methacrylic acid and 4mmol 4-vinylpyridine into a 250mL Erlenmeyer flask, and then add 15mL chloroform. Measure 150 mL of distilled water, add 6.0 mL of sodium lauryl sulfate (according to 4% of the volume of distilled water), heat to completely dissolve, and cool to room temperature. Distilled water was poured into a conical flask, and a crosslinking agent ethylene glycol dimethacrylate (40 mmol, 7.56 mL) and an initiator azobutadiisodinitrile (200 mg) were added. Ultrasonic degassing for 2 minutes, emulsification with a dispersing emulsifier for 5 minutes, and polymerization in a water bath at 60°C for 12 hours. The obtained polymer was washed with distilled water until clarified, and then ultrasonically eluted with 50% acetonitrile-ammonium acetate buffer as the eluent. Finally, the solid-phase extraction device was washed with 60% acetonitril...

Abstract

The invention provides a molecular imprinted polymer microsphere for fast detecting malachite green in aquatic products, which is prepared by mixing two functional monomers at a certain ratio, mixing a malachite green template with the monitor at a certain ratio, sequentially adding a crosslinking agent, an initiator, an emulsifying agent and a lipophilic solvent, and preparing the molecular imprinted polymer microsphere. The invention further provides a method for detecting the malachite green in a sample, which comprises the following steps of: identifying, adsorbing and concentrating a processed aquatic product, and determining the concentration of the malachite green by spectrophotometry. Additionally, the sample can be concentrated to lower the detection limit, so that the method is especially suitable for detection of low-content samples.

Description

technical field [0001] The invention relates to a molecular imprinting detection technology, in particular to a method for rapidly detecting malachite green in aquatic products using the molecular imprinting technology, and the invention also relates to molecular imprinted polymer microspheres used in the detection method. Background technique [0002] As we all know, malachite green has good antibacterial effect in aquaculture, and its price is low. At present, the publicity of its harmfulness is weak, which makes it widely used by many aquaculture households. Based on this, my country has formulated the standard "Limits of Fishery Drug Residues in Pollution-Free Food and Aquatic Products (NY5070-2002)", which stipulates that no harmful substances such as malachite green should be detected in aquatic products. [0003] The 2000 China Export Monitoring Plan for Toxic and Hazardous Substance Residues in Foods of Animal Origin included the monitoring of malachite green residue...
