Embodiment 1

[0033] Example 1: Antibody-modified polydopamine-coated gold nanorod material synthesis

[0034] At room temperature, mix 5 mL of chloroauric acid solution (0.5 mM) with 5 mL of CTAB solution (0.2 M) thoroughly, then add 0.6 mL of pre-cooled sodium borohydride solution (0.01 M), and react in the dark at 28°C for 2 hours to obtain Brown gold nanoseed solution. Mix 5mL chloroauric acid solution (1 mM) with 5mL CTAB solution (0.2 M) thoroughly, add 0.12mL silver nitrate solution (10mM) and mix well, add 0.055mL ascorbic acid solution (0.1M, ready-to-use) and mix well Mix well, add 0.012mL of the gold nano-rod solution synthesized in step (1), mix well and react overnight at 28°C to obtain a gold nanorod material with an aspect ratio of 3.8, centrifuge at 8000rpm, and wash with water twice. Redissolve the washed gold nanorod material in Tris buffer solution (10mM, pH8.0), add 0.01mL p-mercaptophenylboronic acid solution (10mM), shake and react for 1 hour, then add 0.05-0.02mL dop...

Embodiment 2

[0035] Example 2: The application of an integrated platform based on polydopamine-coated gold nanorods for tumor cell Raman imaging detection and real-time photothermal therapy in Raman imaging and photothermal therapy of prostate cancer cells (DU145).

[0036] First add about 10 5 / mL cells, cultured overnight to allow the cells to adhere to the wall. Take 10-30 μL of antibody-modified polydopamine-coated gold nanorod solution, add it to a petri dish, and incubate with slight shaking at 37°C for 1-2h, discard the upper medium, wash with PBS three times, and add 100 uL of PBS.

[0037] Place the glass-bottom culture dish on the Raman microscope stage, adjust the focal length so that the cells can be clearly displayed in the field of view, select the appropriate excitation light (785nm) and adjust the corresponding test parameters, and use the Raman microscope point scanning method Perform rapid Raman scanning on the cells, using the characteristic Raman signal peak of p-merc...

Abstract

The invention belongs to the technical field of biology, and in particular relates to a polydopamine coated gold nanorod material as well as a preparation method and application of the polydopamine coated gold nanorod material. The preparation method disclosed by the invention comprises the following steps: by taking CTAB (cetyl trimethyl ammonium bromide) as a template agent, synthesizing a polydopamine coated gold nanorod material; modifying an antibody onto the surface of the material, thereby obtaining a multifunctional polydopamine coated gold nanorod material. Thus, a tumor cell Raman imaging detection and real-time photothermal therapy platform is constructed. The experiments prove that the gold nanorod material is capable of realizing specific effective tumor target cell marking and performing rapid surface-enhanced Raman spectral imaging scanning on the tumor cells under excitation of near-infrared light; and the tumor target cells are accurately identified by the imaging results, and after the target cells are identified, the infrared light can be focused onto the tumor target cells, the irradiation time is prolonged, and the material is heated by utilizing the photo-thermal conversion effect of the gold nanorod material so as to kill the tumor cells in real time. The polydopamine coated gold nanorod material disclosed by the invention is novel, convenient, practical and efficient and has huge clinical application potential.

Description

technical field [0001] The invention belongs to the field of biotechnology, and in particular relates to a polydopamine-coated gold nanorod material and a preparation method thereof, as well as an application in constructing a tumor cell Raman imaging detection and real-time photothermal treatment platform. Background technique [0002] Imaging guidance is a rapidly developing technology for tumor cell detection and treatment, and is an important means to achieve precision medicine in the future. Among imaging-guided tumor therapy techniques, photothermal therapy has received more and more attention due to its advantages of high efficiency, minimal invasiveness, and low damage to surrounding tissues. In recent years, with the continuous synthesis of a variety of multifunctional nanomaterials with both photothermal conversion efficiency and high imaging sensitivity, people have developed a variety of different imaging-guided photothermal therapy technologies for tumors.
