Embodiment 1

[0023] Preparation of oxygen sensor

[0024] (1) Prepare 200 mL of 5 wt % anhydrous sodium carbonate solution, boil it, put 6.0 g silkworm cocoon layer into it for the first degumming for 30 minutes, and dry the obtained incompletely degummed silk protein at 55° C. for 1 hour.

[0025] (2) Carry out the second complete degumming treatment according to the method of the first degumming to obtain completely degummed silk protein, and dry at 55° C. for 1 hour for later use.

[0026] (3) Prepare 40ml of 50wt% anhydrous calcium chloride solution, boil, put in completely degummed dry silk fibroin, dissolve, boil for 5min, and make completely degummed silk fibroin become a solution.

[0027] (4) Transfer the obtained solution to a dialysis bag, and perform running water dialysis for 72 hours. Finally, the obtained silk protein solution is concentrated to obtain a volume concentration of 7.0% silk protein solution. The silk protein solution is detected as follows: figure 1 Middle cur...

Embodiment 2

[0030] draw standard curve

[0031] (1) Place the oxygen sensor in nitrogen-oxygen mixed gas with oxygen content of 10%, 20%, 30%, 40%, and 50%, take it out after 0.5h, and obtain the reflection intensity and oxygen content by using a USB4000 fiber optic spectrometer The relationship, the standard curve such as image 3 , see the spectrogram Figure 4 .

Embodiment 3

[0033] Pass the air to be detected into the closed space where the oxygen sensor prepared in Example 1 is placed, continue to pass through for 0.5 hours, and detect it with a USB4000 fiber optic spectrometer. The detection diagram is as follows Figure 5 Shown, calculate the oxygen content in the gas according to the standard curve, and the calculation result is: 21.3%, (21.7% of the oxygen content in the actual air), illustrating that the method error of the present invention's detection oxygen is little, and the result is accurate.

Abstract

The invention relates to a method of detecting the oxygen content and in particular relates to a method of quickly detecting the oxygen content in gas by using a fibroin membrane, and belongs to the optical field. According to the technical scheme provided by the invention, the method specifically comprises the following steps: degumming a silkworm cocoon layer twice and performing primary salt dissolving, performing dialysis by flowing water for 72h, mixing the dialyzed silkworm cocoon layer with hemoglobin, dropwise adding the mixture onto a polycarbonate sheet after film removal, and drying the polycarbonate sheet in an oven to form a film to obtain an oxygen sensor; and after full contact of the oxygen sensor and to-be-detected gas, performing detection by virtue of a fiber optic spectrometer to obtain the oxygen content. By virtue of the characteristic that the hemoglobin carries oxygen, the hemoglobin is fixed to the fibroin to measure the oxygen content in air, and the method is simple and accurate to measure. Moreover, the used materials are biological materials which are green and environmental-friendly, are relatively long in service life, relatively huge in application value and relatively good in prospect.

Description

technical field [0001] The invention relates to the field of optics, in particular to a method for quickly detecting the oxygen content in gas by using a reflection spectrometer to measure the reflection of a silk protein film containing hemoglobin. Background technique [0002] Silk protein is a protein extracted from silk fiber. It is non-toxic, non-irritating, and has good air and moisture permeability, excellent biocompatibility and biodegradability. In the field of biotechnology, silk protein membranes can be used as carriers for immobilized enzymes and for the preparation of biosensors. [0003] Hemoglobin exists in the red blood cells of the blood, and a large amount of hemoglobin is produced during the maturation of red blood cells and can be combined with oxygen. There are many methods for detecting oxygen content in the prior art, mainly including chemical method, electrochemical method, zirconia concentration cell method, yellow phosphorus luminescence method, co...
