Abstract

The invention relates to a method for extracting a heparin-like compound from a swimming bladder. The method comprises the following steps: S1, drying and crushing the swimming bladder, and adding a sodium chloride solution and protease to perform enzymolysis, wherein the material liquid ratio is 1:(20-35), the pH value is 7.5 to 9, the temperature is 45 to 60 DEG C and the enzymolysis time is 4 to 8 hours; S2, performing enzyme deactivation and centrifugation on the enzymolysis liquid obtained in the step S1, taking the supernatant, passing through an anion exchange resin to adsorb, eluting with a sodium chloride solution with the molar fraction being 0.7 to 0.9 mol / L for the first time, and eluting with a sodium chloride solution with the molar fraction being 1 to 1.5 mol / L for the second time; and S3, performing ultrafiltration on the eluent obtained by elution for the second time, and then performing alcohol precipitation and freeze drying to obtain the heparin-like compound, wherein the protease comprises alkaline protease with the mass fraction being 2 to 5 percent and bromelain with the mass fraction being 1 to 4 percent. The heparin-like compound extracted by the method provided by the invention has high purity and content up to above 85 percent, and has a good separation effect with other glycosaminoglycan.

Description

technical field [0001] The invention relates to the technical field of extracting active polysaccharides, in particular to a method for extracting heparin-like compounds from swim bladders. Background technique [0002] my country is a big country in the production, trade and consumption of aquatic products. With the continuous development of the fishery and aquatic product breeding industry, the output of aquatic products in my country has increased from 4.497 million tons to 69 million tons from 1980 to 2016, accounting for more than one-third of the global aquatic product output. At the same time, the aquatic product processing industry is also developing rapidly, and a large amount of processing waste is produced every year, which not only causes a waste of fishery resources, but also causes serious pollution to the environment. Among them, swim bladder is a valuable resource for both medicine and food, rich in active ingredients, and worthy of full development and util...
