Abstract

The invention discloses a method for separating and preparing gelsemium alkaloid from gelsemium elegans benth by virtue of an ion exchange method. The method comprises the following steps: 1) heatingand conducting reflux extraction on a medicinal material, namely the separating and preparing gelsemium alkaloid from gelsemium elegans benth by virtue of ion exchange method, so that a coarse extracting solution is obtained; adding hydrochloric acid to the coarse extracting solution and regulating pH value to 2-5, and conducting filtering; dissolving a precipitate by virtue of a hydrochloric acidwater solution which is 2-5 in pH value, implementing filtering, and combining filtrates so as to prepare a loading solution which is 0.75-1.5g crude drug / mL in concentration; processing the loadingsolution by virtue of a pre-processed strong acid ion exchange resin column, wherein the flow rate of the loading solution is 1-5BV / h; after conducting exchange, rinsing the resin column to be neutral, removing impurities by virtue of ethanol which is 80% in volume concentration, and eluting the resin column by virtue of ammonium hydroxide which is 50% in volume concentration and ethanol which is90% in volume concentration; and collecting eluents, implementing decompressing and recovering the ethanol, and conducting drying, so that a gelsemium alkaloid extract is obtained. With the application of the method provided by the invention, the content of koumine is improved from 4.6% of the coarse extracting solution to be greater than 20% of the coarse extracting solution, so that a foundationis laid for further separation and preparation of the high-purity koumine.
