Embodiment 1

[0096] Example 1, HbHAK1 can promote the absorption of potassium ions by Arabidopsis under low potassium conditions

[0097] This example provides a protein derived from wild barley (Hordeum brevisubulatum), which can promote the absorption of potassium ions under low potassium conditions. Its name is HbHAK1, its amino acid sequence is sequence 1 in the sequence listing, and its CDS sequence is the sequence listing Medium sequence 2.

[0098] 1. HbHAK1 can promote the absorption of potassium ions in Arabidopsis under low potassium conditions

[0099] The steps of HbHAK1 function detection are as follows:

[0100] 1. Construction of recombinant vector

[0101] The pCambia1302 vector (Zhang H, Zhao FG, Tang RJ, Yu Y, Song J, Wang Y, Li L, Luan S. (2017) Two tonoplast MATE proteins function as turgor-regulating chloride channels in Arabidopsis.Proc Natl Acad Sci U S A .114:E2036-E2045) The DNA fragment between the BglII and SpeI recognition sequences is replaced by the HbHAK1 co...

Embodiment 2

[0179] Embodiment 2, HbHAK1 gene expression analysis

[0180] When the wild barley seedlings were cultivated to two leaves and one heart, under the stress conditions of NaCl, ABA, mannitol and PEG6000, the root materials were taken at each time point of 0, 30min, 1hr, 3hr, 6hr and 12hr, and the genes were analyzed by real-time fluorescent quantitative PCR Express the situation. Each stress treatment was carried out by adding corresponding substances to 1 / 2 Hoagland culture medium. The concentrations of NaCl, ABA, mannitol and PEG6000 in the stress-treated culture medium were 350mM, 50μM, 350mM and 15mg / 100mL, respectively. 1 / 2 Hoagland's culture solution is a culture solution obtained by halving the concentration of each solute in Hoagland's nutrient solution. Primers and internal references used in real-time fluorescent quantitative PCR are the same as in Example 1.

[0181] The induced expression pattern of HbHAK1 gene in roots under different abiotic stresses was determin...

Abstract

The invention discloses protein for promoting efficient absorption of potassium ions and a coding gene thereof. The protein for promoting efficient absorption of the potassium ions disclosed by the invention comprises the following protein A1, A2 or A3: A1, protein with the amino acid sequence being sequence 1; A2, proteins with the amino acid sequence shown as the sequence 1 in a sequence table subjected to substitution and / or deletion and / or addition by one or several amino acid residues, and with identical functions; A3, fused protein obtained by connecting labels at the N end and / or C endof the A1 or A2. When the protein provided by the invention is expressed in plants, the potassium ion absorption capability and the salt tolerance of the plants can be improved; compared with HAK family genes of the other plants, the protein shows high potassium absorption capability under the low-potassium condition. The result shows that the protein and the coding gene of the protein disclosed by the invention can promote the efficient absorption of the potassium ions and have wide application prospects.

Description

technical field [0001] The invention relates to a protein and its coding gene for promoting efficient absorption of potassium ions in the field of biotechnology. Background technique [0002] At present, the lack of molecular mechanisms of plant salt-alkali tolerance and drought tolerance has become a bottleneck problem in the development of modern agriculture, which has seriously hindered the development of improved crop breeding. It is of great significance to use plant resources to cultivate drought-resistant and saline-alkali-resistant green plants to improve the environment and increase crop yields. Maintaining potassium and sodium balance in plants plays a pivotal role in improving plant salt tolerance. Understanding the molecular mechanism of plants' efficient absorption and utilization of potassium under salt stress has broad application value in agriculture and environmental improvement. Potassium is the most abundant cation in the cytoplasm, accounting for 10% of ...
