Embodiment 1

[0017] Streptomyces GSDX-1318 can grow at 28-37°C and pH is slightly alkaline. The hyphae extend radially, forming uniaxial spores with regular helices, and the helix is tight. The hyphae are darker in color and thicker in diameter. Most of the aerial hyphae mature and differentiate into sporocydus, tightly spiraled uniaxial spore heads. Clusters of conidia are produced by septal division. Streptomyces GSDX-1318 was inoculated on a variety of commonly used natural and synthetic media for culture, and the culture characteristics were observed (Table 1).

Embodiment 2

[0021] Inoculate Streptomyces sp.GSDX-1318 on a nutrient medium with 0.5% soluble starch, 0.4% yeast extract powder, 0.4% glucose, 0.4% soybean peptone, 2% agar, the rest of water, and pH 7.2. For cultivation, the cultivation temperature is 28° C., and the cultivation time is 3 days. Take the slant bacteria, add 5ml of normal saline (per 18×180mm test tube slant bacteria), gently wash the bacteria lawn with an inoculation needle to make a spore suspension, take 2ml of the spore suspension and inoculate it into 100ml of the seed medium Cultivate for 2 days, the seed medium contains 2.0% mannitol, 4.0% soybean meal powder, 3% L-valine, the balance of water, pH 7.2, the culture temperature is 28° C., and the rotation speed is 220 rpm. Then, 5ml of seed liquid was inserted into a 500ml shake flask with a liquid content of 100ml of fermentation medium, and fermented on a shaking table at 28°C, 200rpm, and pH7.2 for 4 days to obtain Streptomyces sp.GSDX-1318 fermentation liquid, th...

Embodiment 3

[0025] Inoculate Streptomyces sp.GSDX-1318 on 0.6% soluble starch, 0.6% yeast extract powder, 0.6% glucose, 0.5% soybean peptone, 1.8% agar, the rest of water, pH 7.2 nutrient medium, and carry out slant culture , the culture temperature was 28°C, and the culture time was 5 days. Take the slant bacteria, add 5ml of normal saline (per 18×180mm test tube slant bacteria), gently wash the bacteria lawn with an inoculation needle to make a spore suspension, take 2ml of the spore suspension and inoculate it into 100ml of the seed medium Cultivate for 3 days, the seed medium contains 3.0% mannitol, 4.0% soybean meal powder, 3% L-valine, the balance of water, pH 7.2, the cultivation temperature is 28° C., and the rotation speed is 220 rpm. Then 5ml of the seed liquid was inserted into a 500ml shake flask with a liquid content of 100ml of fermentation medium, and fermented on a shaking table at 28°C, 200rpm, and pH 7.2 for 6 days to obtain Streptomyces sp.GSDX-1318 fermentation broth ...

Abstract

The invention discloses an avilamycin producing strain which is Streptomyces sp. GSDX-1318. The systematic nomenclature and the Latin scientific name are Streptomyces sp.. The invention also provides a fermentation method for producing oligosaccharide antibiotic, namely avilamycin. The method comprises the following steps of: (1) performing slant culture, namely inoculating the Streptomyces sp. GSDX-1318 into a nutrient culture medium and performing slant culture; (2) performing seed culture on spores obtained through slant culture to obtain seed liquid; and (3) performing fermentation culture, namely inoculating the seed liquid into a fermentation culture medium for culturing to obtain zymotic fluid of the Streptomyces sp. GSDX-1318, and separating to obtain the oligosaccharide antibiotic, namely the avilamycin. By the fermentation method for producing the oligosaccharide antibiotic, namely the avilamycin, the process is simple and production cost is low. The oligosaccharide antibiotic, namely the avilamycin produced by the fermentation method can be taken as a novel broad-spectrum, high-efficiency and safe feed additive.

Description

technical field [0001] The invention relates to streptomyces and a method for fermenting and producing oligosaccharide antibiotic avelamycin, belonging to the fields of agricultural biotechnology, industrial biotechnology and fermentation engineering. Background technique [0002] It has been nearly 60 years since antibiotics were added to feed to promote the growth and production of livestock and poultry, and many countries have used antibiotic additives for a long time. However, with the widespread use of antibiotic growth promoters, a series of problems have gradually emerged, mainly drug residues and drug resistance. The long-term, extensive and indiscriminate use of antibiotic feed has brought great trouble and difficulty to the control and treatment of bacterial diseases, and also brought potential threats to human health. Although there are many controversies about whether antibiotics should be used in feed, practice has proved that rational use of antibiotic additiv...
